Synthesis and Characterization of Nanofiber of Gelatin, Polyvinyl Alcohol, and Chitosan for Wound Dressing Application

Jul 16, 2026

Electrospun nanofiber formulations based on gelatin, polyvinyl alcohol, and chitosan were synthesized, including gelatin/PVA and gelatin/PVA/chitosan blends. Morphology and structure were evaluated by SEM and FTIR, while drug-loaded fibers containing ciprofloxacin and ibuprofen were characterized by FTIR, SEM, XRD, and DSC. SEM indicated homogeneous systems, and FTIR showed functional-group interactions; ciprofloxacin-loaded nanofibers exhibited antibacterial activity against Escherichia coli and Staphylococcus aureus in vitro.

RESEARCH ARTICLE Synthesis and characterization of nanofiber of gelatin, polyvinyl alcohol, and chitosan for wound dressing application Padideh Aliaghaa, Sahar Amiria and Majid Ghiassb aDepartment of Polymer Engineering, Science and Research Branch, Islamic Azad University, Tehran, Iran; bDepartment of Polymer Science, Faculty of Science, Iran Polymer and Petrochemical Institute, IPPI, Tehran, Iran ABSTRACT Different formulations of electrospun nanofiber were produced from gelatin, chitosan, polyvinyl alcohol, gelatin/polyvinyl alcohol, and gelatin/polyvinyl alcohol/chitosan. SEM and FTIR were used to determine the morphology and structure of the obtained nanofiber. In the following steps, ciprofloxacin and ibuprofen were loaded into the obtained nanofiber, which was then characterized with FTIR, SEM, XRD, and DSC. The SEM morphological characterization revealed that the systems were homogeneous. FTIR analysis of the component’s structure revealed interactions among some functional groups. In ciprofloxacin-containing nanofibers demonstrated significant antibacterial activity against Escherichia coli and Staphylococcus aureus in vitro, which is dependent on the antibacterial activity of both ciprofloxacin and chitosan. ARTICLE HISTORY Received 23 August 2022 Accepted 16 February 2023 KEYWORDS Chitosan; nanofiber; gelatin; polyvinyl alcohol; antibacterial activity; electro-spinning Introduction One of the challenges that medical science has faced for along time is offering new methods for treating damaged tissues as wounds on the largest human organ, the skin (Rezvani Ghomi et al., 2019) . As a result of thermal or physical damage to the skin, an injury results in a disintegration of the skin’s defensive function (Dhivya et al., 2015; Xinkuan et al., 2021) . The role of a wound dressing is to provide the best conditions for wound healing while protecting the wound from further invasion by microorganism (Carolina et al., 2021) . The entry and growth of germs in the wound may lead to infection, and if the wound does not heal completely after 48h, it will probably become infected due to the entry of dust and germs that remain in the wound. There are several symptoms associated with infection, including high fever and fatigue, severe pain and burning, swellings and redness surrounding the injury, and most importantly, tetanus infection in deep or dirty wounds (Bakhsheshi-Rad et al., 2017; Makwena et al., 2021; Mohamed et al., 2022; Rezvani Ghomi et al., 2019; Sung Giu, 2022) . Wounds are used as abed for wound healing. The primary role of such a bed is to maintain skin moisture and minimize wound damage, and barrier to the entry of external factor (More et al., 2021) . Various antibacterial medications such as ciprofloxacin and ibuprofen are used to prevent and treat wound infection; they also eliminate fever and analgesia, especially in inflammatory pai (Sahoo et al., 2011) . Ciprofloxacin (CIP) is a common antimicrobial dru
