Objective: investigate how hsa_circ_0075451 promotes NSCLC proliferation, metastasis, and glycolysis through interaction with the RNA-binding protein RBM4. Methods: NSCLC and paracancer tissues from 53 cases were analyzed by RT-qPCR and Western blot; sequences/plasmids were transfected into A549 cells and malignant phenotypes were assessed with CCK-8, EdU, Annexin V-PI, and Transwell. Glycolysis was measured by glucose use, lactate production, and glycolysis-related proteins; in vivo tumorigenesis and mechanistic assays verified the hsa_circ_0075451–RBM4 interaction. Results: hsa_circ_0075451 was elevated in NSCLC, and knockdown reduced proliferation, metastasis, glycolysis, and tumor growth; RBM4 mediated these effects.

Results Hsa_circ_0075451 levels were elevated in NSCLC, and reducing its expression diminished cell proliferation, metastasis, glycolysis, and tumor expansion. Hsa_circ_0075451 interacted with RBM4 . RBM4 down-regulation restricted NSCLC cell proliferation, metastasis, and glycolysis. RBM4 up-regulation opposed the effects of hsa_circ_0075451 knockdown. Conclusion Hsa_circ_0075451 promotes NSCLC proliferation, metastasis and glycolysis by interacting with RBM4 .
