Cancer Vaccine Development: Toward Artificial Intelligence-Assisted Personalized Cell Membrane Nanovaccine

Sep 22, 2026

Cancer vaccines offer a promising immunotherapeutic strategy, yet many clinical candidates fail to deliver statistically significant survival benefits. Nanotechnology-enabled cancer nanovaccine platforms address these shortcomings by enhancing antigen delivery efficiency and strengthening antitumor immune responses. Cell membrane nanoparticle (CNP)-based vaccines are especially studied due to their strong biocompatibility and immunogenicity. Maintaining major histocompatibility complex (MHC)-associated peptide repertoires on CNPs can support endogenous antigen presentation while reducing dependence on in silico tumor antigen prediction.

However, most candidates have not achieved statistically significant improvements in patient survival. To address these limitations, nanotechnology-enabled cancer nanovaccine platforms have attracted increasing attention. Relative to conventional vaccine formats, cancer nanovaccines can improve antigen delivery efficiency and promote robust antitumor immune responses. Among these approaches, cell membrane nanoparticle (CNP)-based vaccines have been widely investigated and show strong biocompatibility and immunogenicity. Notably, preservation of major histocompatibility complex (MHC)-associated peptide repertoires on CNPs can help leverage endogenous antigen presentation and may reduce reliance on in silico tumor antigen Abbreviations: 201M, gp100:209-217; AAM, Antibody-anchored membrane; AI, Artificial intelligence; AI-cell, Artificial Immune Cell; AiLNP, Artificial ICD LNP; APC, Antigen-presenting cell; ApoA1, Apolipoprotein A1; ATRA, All-trans-retinoic acid; BBB, Blood-brain barrier; bIL-15, Membrane bound IL-15/IL-15 receptor α complex; CaP, Calcium phosphate; CCM, Cancer cell membrane; CCR, C-C motif chemokine receptor; CD, Cluster of differentiation; CDX-110, Rindopepimut; cGAS, Cyclic GMP-AMP synthase; circRNA, Circular RNA; CLX, Celecoxib; CNN, Convolution Neural Network; CNP, Cell membrane nanoparticle; COMET, Composite Material Transformer; COX-2, Cyclooxygenease-2; CRT, Calreticulin; CTL, Cytotoxic T lymphocyte; CVB3, Coxsackievirus B3; CXCR4, C-X-C motif chemokine receptor 4; DAMP, Damage-associated molecular pattern; DC, Dendritic cell; dLN, Draining lymph node; DNN, Deep Neural Network; DOX, Doxorubicin; DP, Diphenylalanine; EBV, Epstein-Barr virus; ECM, Extracellular matrix; EGRFvIII, Epidermal growth factor receptor variant III; ER, Endoplasmic reticulum; ESC, Embryonic stem cell; ESO, Esomeprazole; FBS, Fetal bovine serum; GA, Gambogic acid; GBM, Glioblastoma; GI, Gastrointestinal; GM-CSF, Granulocyte-macrophage colony-stimulating factor; GNN, Graph Neural Network; gp100, Glycoprotein 100; GSH, Glutathione; HDL, High-density lipoprotein; HER2, Human epidermal growth factor receptor 2; HMGB1, High-mobility group box 1; HNSCC, Head and neck squamous cell carcinoma; ICB, Immune checkpoint blockade; ICD, Immunogenic cell death; IDO, Indoleamine 2,3-dioxygenase; IFN, Interferon; IL, Interleukin; iNKT, Invariant natural killer T cell; irAE, Immune-related adverse effect; IV, Intravenous; IVIS, In vivo imaging system; JAK, Janus kinase; KO, Knockout; LiON, Likelihood-based Optimization of Nanoparticles; LLM, Large Language Model; LN, Lymph node; LNP, Lipid nanoparticle; LSTM, Long Short-Term Memory; LSTV, Large-scale structural tumor variant; M, Microfold cell; mAgs, Membrane-associated antigens; MDSC, Myeloid-derived suppressor cell; MHC, Major histocompatibility complex; MLP, Multi-Layer Perceptron; MMP, Metalloproteinase; Mn-LDH, Mn-based layered double hydroxide; MOF, Metal-organic framework; MPI, Polybia-mastoparan O; MPLA, Monophosphoryl lipid A; MSN, Mesoporous silica nanoparticle; NGS, Next-generation sequencing; NIR, Near-infrared; NK, Natural killer cell; NLRP3, NOD-like receptor pyrin domain-containing pr
