The present study evaluates the bioaccessibility and metabolic transport of different dried thinned peach polyphenols using in vitro simulated digestion and Caco-2 cell models. Compared with freeze drying (FD) and natural drying (ND), hot air drying at 50 °C (HAD50) produced the highest simulated gastrointestinal bioaccessibility (91.19%) and favorable absorption in Caco-2 cells. Co-incubation revealed a combined transepithelial transport effect, 26 metabolites were detected, and P-gp and OATP transport mechanisms modulated absorption.

The present study demonstrates that the HAD50 treatment of thinned peaches is proved to be an effective way to enhance the bioaccessibility and permeability of polyphenols compared to FD and ND. | 1. Introduction Thinned peaches are the superfluous growing fruits that are thinned out at approximately a month after the full-bloom stage, and they are characterized by a high abundancy of bioactive compounds such as polyphenols, amino acids and organic acids. In particular, the polyphenol content in thinned peaches are approximately 8 times higher than the corresponding ripe fruit (Guo, Bi, Li, Lyu, Wu, & Xu, 2020). Many health benefits attributed to fruits consumption are correlated with the presence of phenolic contents (Makhtoomi et al., 2024). Furthermore, a number of in vitro and in vivo trial-based reports have exhibited that increasing dietary consumption of polyphenols can reduce the incidence of chronic diseases such as diabetes, stroke, cardiovascular disease and cancer (Bohn, 2014; He et al., 2017). However, the bioaccessibility and metabolic transformation of polyphenols during digestion is a key aspect to their biological effects (He et al., 2017; Yinet al., 2022). In this regard, Brown et al. (2014) have previously demonstrated that bilberry polyphenols have a large modification tendency during in vitro and in vivo ileal fluid digestion, and both simulated extracts showed long-lasting biological activity in an anti-genotoxicity model. Approximately 1.25 million tons of the thinned peaches is manually or mechanically removed from the trees to ensure the remaining peaches achieve marketable size, quality, and sugar content (Guo, Bi, Li, Lyu, Zhou, & Wu, 2020). The conversion of thinned peaches into a stable, value-added product is a prime example of agricultural by-product valorisation.
