Here, we show that chronic UVB exposure after melanoma metastasis to the lung inhibits metastatic growth and modestly improves survival in mice. In UK Biobank participants, higher solar radiation exposure is positively associated with improved lung cancer survival. Mechanistically, UVB modulates the tumor immune environment: Mass cytometry of CD45+ immune infiltrate cells from lung metastases of mice UVB-and sham-irradiated revealed a UVB-induced increase inCXCR2− neutrophils and a reduction in Ly6C+ inflammatory monocytes, with no changes in regulatory T cells or expression of immune checkpoint molecules. Together, these results highlight a potentially protective role of UVB and solar radiation and provide a rationale for further investigation into UV as a factor that enhances antitumor immunity. \nSun exposure and UVB radiation exert complex effects on the skin and on internal organs and body systems1. In the context of cancer, UVB is well-established as a mutagen; however, evidence indicates that UVB impacts cancer cells directly and also modulates the immune response to cancer2. Most studies have focused on the direct effects of UVB on the immune response within the skin. The influence of UVB on the immune response to primary cancers of internal organs and metastases remains poorly understood. \nChronic exposure to UVB radiation clearly has suppressive effects on the skin immune system. For example, chronic UVB exposure suppresses prototypic T cell-mediated contact hypersensitivity to chemical haptens3 and delayed-type hypersensitivity to viral, bacterial, and fungal antigens4–6. UVB influences the immune system through multiple mechanisms: It reduces the number, function, and morphology of the Langerhans cells7 and impairs their ability to present antigen to effector CD4+ T helper type 1 cells8. UVB also promotes skin inflammation by inducing synthesis of inflammatory cytokines IL-1, IL-6, IL-8, TNFα, and type I IFN by skin keratinocytes and resident leukocytes8,9. In turn, these cytokines mediate chemotactic signaling in neutrophils and macrophages in the skin7,9. The skin immune-suppressive effect of UVB is the reason for its efficacy as a treatment for skin-related autoimmune diseases including psoriasis and vitiligo10, 11. \nThe immunosuppressive and inflammatory effects of UVB are also reflected in the context of the immune system’s response to skin cancer. When mice are exposed to UVB prior to tumor injection, the immunosuppressive effects of UVB decrease the local tumor immune response, resulting in enhancement of melanoma tumor growth12. Recently, we showed that chronic UVB exposure prior to tumor injection induces resistance to antiPD-1 immunotherapy and anti-Ly6a antibody treatment; Ly6a is upregulated on T cells following UVB13.
