Metastasis—the process by which malignant cells spread from the primary tumor to distant sites—is a defining characteristic of cancer and the leading cause of cancerrelated mortality (2) . Metastatic progression requires the travel of cancer cells and their adaptation and constant evolution through continuous and complex interactions with their surrounding niches, eventually allowing them to change their normal behavior and become aggressive (3) . Despite the critical role of metastasis in cancer’s lethality, its underlying mechanisms are still not fully understood. \nThe epithelial–mesenchymal transition (EMT) is a biological process in which epithelial cells lose their original characteristics and acquire mesenchymal properties (4) . This transition enhances the cells’ ability to move and invade, thereby contributing to metastatic spread (4). As tumor cells undergo EMT, they gain the ability to invade nearby tissues and enter circulation, enabling dissemination to distant organs (5) . Intriguingly, once they reach a new site, these cells may undergo a reverse process, known as the \n1Tissue and Tumor Microenvironments Lab, Cancer Discovery and Regenerative Medicine Program, Lee Kong Chian School of Medicine, Nanyang Technological University, Singapore, Singapore. 2Lee Kong Chian School of Medicine, Nanyang Technological University, Singapore, Singapore. 3Multidisciplinary Institute of Ageing (MIA-Portugal), Coimbra, Po","cbCais7bhDHjTAsJ","https://ap.wps.com/l/cbCais7bhDHjTAsJ","pdf",8194261,13,"English","# Introduction\n## EMT and metastatic progression\n## Tumor microenvironment composition\n## Rationale for targeting vascular and stromal compartments\n## Scope of the review","[{\"question\":\"What role does the tumor microenvironment (TME) play in cancer progression?\",\"answer\":\"The TME fosters interactions between cancer cells and surrounding cellular and acellular elements, supporting tumor growth, metastatic potential, and resistance to therapies.\"},{\"question\":\"How are epithelial–mesenchymal transition (EMT) and metastasis connected?\",\"answer\":\"EMT enables cells to move and invade, supporting metastatic spread by allowing tumor cells to disseminate to distant organs; a reverse process can further support colonization and growth.\"},{\"question\":\"Why are vascular and stromal cells considered promising therapeutic targets?\",\"answer\":\"Vascular and stromal cells contribute to EMT regulation and resistance pathways, and targeting their components is proposed as a promising therapeutic approach.\"}]","Transforming Cancer Therapy - Targeting Vascular and Stromal Cells in the Tumor Microenvironment | PDF",1790189576,33]
