We introduce four related signaling pathways, namely cAMP/PKA/CREB signaling way, HIFα signaling way, NF-κB signaling way, STAT3 signaling way, FOXO signaling pathway and Hedgehog (HH) signaling pathway. We also emphasize the current understanding of the complex roles CRY1 plays in the progression of various solid tumors. We propose that its function represents a novel therapeutic target that could pave the way for new cancer treatments. Additionally, we summarize three emerging therapeutic strategies based on the roles of CRY1: targeting CRY1 with small molecules, using CRY1 as diagnostic and therapeutic tools, and promoting drug efficacy through CRY1 modulations. It is imperative to underscore the significance of conducting more extensive research to translate the insights obtained from cellular and animal models into practical human applications, ultimately enhancing therapeutic outcomes. This is accomplished through clock gene oscillations, which govern the daily activities of the majority of light-sensitive species, including humans and cyanobacteria [2]. The core clock genes identified in humans include Cryptochrome-1 (CRY1), which is the focus of our review [3]. Cryptochrome (CRY) was initially identified as a potential photoreceptor in Arabidopsis thaliana. It was clonedas the gene that compensates for the hy4 photomorphogenic mutant, which exhibits a defect in the blue-lightdependent suppression of hypocotyl elongation [4]. In birds and insects, photoexcited cryptochrome has a role in magnetoreception [7]. \nCancer development arises from cell overproliferation and genetic instability [8–10]. Historically, cancer rarely contributed to mortality due to the shorter average life expectancy. However, advancements in medical care over the past century have greatly enhanced life expectancy. As a consequence, the total incidence of cancer has increased, resulting in a significantly higher number of cases. According to the World Health Organization (WHO), one in six deaths globally are due to cancer, making it the second most common cause of death [11]. \nVarious studies have revealed the connection between CRY1 and tumors, including the environmental disruption of circadian ryhthms, polymorphisms (SNPs) and other related elements, which offers valuable understanding into the possible therapeutic applications of CRY1 \n© The Author(s) 2025.
