STM2457, a highly efficient METTL3 inhibitor, can inhibit METTL3 activity and may serve as a potential therapeutic strategy in cancers. However, the role of STM2457 for GC cells is still unknown. In this study, we analyzed the expression profile data of GC in TCGA and GEO databases, and further explored the expression involvement of METTL3 in GC cell line, investigated the therapeutic effect of STM2457 targeted inhibition of METTL3 in GC both in vitro and in vivo experiments. The results indicated that STM2457 could suppress GC cell proliferation and migration by inhibiting METTL3, and also promoted cell apoptosis and arrest the cell cycle in S phase. In addition, STM2457 could inhibit tumor growth in subcutaneous xenotransplantation mouse model. Our findings suggested that STM2457 had great potential for the treatment of GC and could serve as a foundation for future clinical applications. \nIntroduction \nGastric cancer is a malignant tumor originating from gastric mucosal epithelium. Its incidence rate ranks fifth and mortality ranks third worldwide [ 1–3] . Due to the difficulty in early detection of gastric cancer, patients in the middle and late stages often experience invasion and metastasis, resulting in poor treatment outcomes and a low 5-year survival rate. Therefore, searching for biomarkers and therapeutic targets \nPLOS One | [https://doi.org/10.1371/journal.pone.0345744](https://doi.org/10.1371/journal.pone.0345744) March 24, 2026 1 / 14 \n202312051358], Natural Science Foundation of Shandong Province [ZR2022MH197], key research and development plan of Jining [2022YXNS152,2023YXNS035], Taishan","cbCaic1a0TX6x2Ro","https://ap.wps.com/l/cbCaic1a0TX6x2Ro","pdf",1549798,14,"English","# Abstract\n# Introduction\n## Gastric cancer burden and need for targets\n## Role of m6A RNA methylation and METTL3 in cancer","[{\"question\":\"What biological pathway does STM2457 target in gastric cancer?\",\"answer\":\"STM2457 inhibits METTL3 activity, thereby modulating N6-methyladenosine (m6A) RNA methylation related processes.\"},{\"question\":\"How does STM2457 affect gastric cancer cell behavior in vitro?\",\"answer\":\"STM2457 suppresses gastric cancer cell proliferation and migration, promotes apoptosis, and arrests the cell cycle at the S phase.\"},{\"question\":\"What evidence supports STM2457 activity in vivo?\",\"answer\":\"STM2457 inhibited tumor growth in a subcutaneous xenotransplantation mouse model.\"}]","The METTL3 inhibitor STM2457 suppresses gastric cancer progression by modulating m6A RNA modification | PDF",1790086712,35]
