Tumor metabolic reprogramming, especially glucose metabolism, is essential for tumor survival. This study examines whether the Hippo pathway co-activator TAZ regulates gluconeogenesis and promotes tumorigenesis in HCC, and whether TAZ acts through the glucocorticoid receptor (GR). Using TCGA and clinical validation, cellular gain/loss of TAZ, gene expression profiling, in vivo tumor models, and mechanistic assays, results identify TAZ upregulation, altered gluconeogenic genes, and a TAZ–GR interaction that suppresses GR transcriptional activity.",{"@graph":14,"@context":77},[15,34,56],{"@type":16
BreadcrumbList",[18,23,27,31
@type":21
https://docshare.wps.com","Home","ListItem",1
@type":21
https://docshare.wps.com/document/","Document",2
@type":21
https://docshare.wps.com/document/healthcare/","Healthcare",3
@type":21
https://docshare.wps.com/document/taz-ww-domain-mediated-regulation-of-gluconeogenesis-and-tumorigenesis-in-hepatocellular-carcinoma-through-interaction-with-the-glucocorticoid-receptor/349159/",4
@type":35
"DigitalDocument
@type":38
https://docshare.wps.com/thumbnails/taz-ww-domain-mediated-regulation-of-gluconeogenesis-and-tumorigenesis-in-hepatocellular-carcinoma-through-interaction-with-the-glucocorticoid-receptor/349159.png","ImageObject",300,407
@type":43},"Đào","Person
@type":46},"DocShare","Organization","application/pdf","2026-10-22","2026-09-22",true,{"@type":52
InteractionCounter",{"@type":54},"ViewAction",6,{"@type":57
FAQPage",[59,65,69,73
@type":61
What problem does the study address in hepatocellular carcinoma?","Question
@type":64},"It addresses poor outcomes in HCC by focusing on how tumor metabolism, particularly glucose metabolism, supports tumor survival and progression.","Answer
@type":61
How was TAZ involvement in HCC evaluated?
@type":64},"TAZ expression was analyzed using TCGA data and validated in clinical samples and cell lines, then TAZ was overexpressed or silenced to test effects on proliferation, apoptosis, migration, and invasion.
@type":61
What is the relationship between TAZ, gluconeogenesis genes, and tumor behavior?
@type":64},"TAZ was significantly upregulated in HCC, and TAZ overexpression increased proliferation while decreasing apoptosis and promoting migration/invasion; Tumor metabolic reprogramming, particularly involving glucose metabolism, is essential for tumor survival. This study investigates the role of the Hippo pathway effector transcriptional co-activator with PDZ-binding motif (TAZ) in regulating gluconeogenesis and promoting tumorigenesis in HCC.
Methods: TAZ expression in HCC was analyzed using The Cancer Genome Atlas data and validated in clinical samples and cell lines. TAZ was overexpressed or silenced in HCC cell lines to evaluate its effects on cell proliferation, apoptosis, migration, and invasion. The expression and prognostic relevance of the gluconeogenesis-related genes phosphoenolpyruvate carboxykinase 1 (PCK1) and glucose-6-phosphatase (G6PC) were examined, along with their correlation with TAZ expression. Tumor growth was assessed in nude mice. Interactions between TAZ and the glucocorticoid receptor (GR) were investigated using co-immunoprecipitation, immunofluorescence, and chromatin immunoprecipitation assays.
Results: TAZ was significantly upregulated in HCC tissues and cell lines. TAZ overexpression enhanced proliferation, reduced apoptosis, and promoted migration and invasion. In contrast, PCK1 and G6PC were downregulated in HCC and showed a negative correlation with TAZ expression.
Conclusion: TAZ modulates gluconeogenesis and accelerates tumor growth, whereas its knockdown attenuates tumor progression. TAZ interacts with GR, suppressing its transcriptional activity on gluconeogenic gene promoters.

| Keywords: Hepatocellular carcinoma; TAZ; Glucose metabolism; Gluconeogenesis genes; Receptors, glucocorticoid | |
| --- | --- |
| Received: 22 May 2025, Revised: 1 September 2025, Accepted: 30 September 2025
Corresponding authors: Li Chen
Department of Oncology, The First Affiliated Hospital, Jiangxi Medical College, Nanchang University, No.17, Yongwai Zhengjie, Donghu District, Nanchang City 330006, Jiangxi Province, China
Tel: +86-13803515919, Fax: +86-0791-7068511, E-mail: [clmedic@126.com](clmedic@126.com)
Yangyang Liu
Department of Oncology, The First Affiliated Hospital, Jiangxi Medical College, Nanchang University, No.17, Yongwai Zhengjie, Donghu District, Nanchang City 330006, Jiangxi Province, China
Tel: +86-15927166267, Fax: +86-0791-7068511, E-mail: [liuyyeyhl@163.com](liuyyeyhl@163.com) | Copyright © 2026 Korean Endocrine Society
This is an Open Access article distributed under the terms of
the Creative Commons Attribution Non-Commercial License ([https://creativecommons.org/licenses/by-nc/4.0/](https://creativecommons.org/licenses/by-nc/4.0/)) which permits unrestricted non-commercial use, distribution, and reproduction in any medium, provided the original work is properly cited.
|

[www.e-enm.org](www.e-enm.org) 267
Huang H, et al.
INTRODUCTION
Hepatocellular carcinoma (HCC) is the most common type of primary liver cancer, ranking sixth in incidence and third in cancer-related m","cbCaiolhkWbKqF0w","https://ap.wps.com/l/cbCaiolhkWbKqF0w","pdf",3159353,21,"English","# Introduction
# Methods
# Results
# Conclusion","[{"question":"What problem does the study address in hepatocellular carcinoma?","answer":"It addresses poor outcomes in HCC by focusing on how tumor metabolism, particularly glucose metabolism, supports tumor survival and progression."},{"question":"How was TAZ involvement in HCC evaluated?","answer":"TAZ expression was analyzed using TCGA data and validated in clinical samples and cell lines, then TAZ was overexpressed or silenced to test effects on proliferation, apoptosis, migration, and invasion."},{"question":"What is the relationship between TAZ, gluconeogenesis genes, and tumor behavior?","answer":"TAZ was significantly upregulated in HCC, and TAZ overexpression increased proliferation while decreasing apoptosis and promoting migration/invasion; gluconeogenesis genes PCK1 and G6PC were downregulated and negatively correlated with TAZ."},{"question":"How does TAZ influence the glucocorticoid receptor in this study?","answer":"The study shows that TAZ interacts with the glucocorticoid receptor and suppresses its transcriptional activity on gluconeogenic gene promoters."}]","TAZ WW Domain-Mediated Regulation of Gluconeogenesis and Tumorigenesis in Hepatocellular Carcinoma through Interaction with the Glucocorticoid Receptor | PDF",1790082013,53]
