B7-H3 is highly expressed in extracranial PSTs, including neuroblastoma, rhabdomyosarcoma, nephroblastoma, osteosarcoma, and Ewing sarcoma, while remaining absent or very low in normal tissues and organs. B7-H3 influences childhood malignant solid neoplasms through immune evasion, tumor invasion, and disruption of cell-cycle regulation. Studies report that B7-H3 knockdown reduces proliferation and migration, suppresses tumor growth, and strengthens anti-tumor immune responses. Antibody-drug conjugates and B7-H3-targeting CAR-T cells show potent preclinical activity, and clinical evidence supports strong anti-tumor effects in metastatic neuroblastoma, with benefits and obstacles discussed across in vitro, in vivo, and clinical research.",{"@graph":69,"@context":123},[70,84,106],{"@type":71,"itemListElement":72},"BreadcrumbList",[73,77,79,82],{"item":74,"name":75,"@type":76,"position":8},"https://docshare.wps.com","Home","ListItem",{"item":78,"name":9,"@type":76,"position":14},"https://docshare.wps.com/document/",{"item":80,"name":35,"@type":76,"position":81},"https://docshare.wps.com/document/healthcare/",3,{"item":83,"name":65,"@type":76,"position":19},"https://docshare.wps.com/document/targeting-b7-h3a-novel-strategy-for-the-design-of-anticancer-agents-for-extracranial-pediatric-solid-tumors-treatment-review/377788/",{"url":83,"name":65,"@type":85,"image":86,"author":91,"headline":65,"publisher":94,"fileFormat":97,"inLanguage":63,"description":67,"dateModified":98,"datePublished":99,"encodingFormat":97,"isAccessibleForFree":100,"interactionStatistic":101},"DigitalDocument",{"url":87,"@type":88,"width":89,"height":90},"https://docshare.wps.com/thumbnails/targeting-b7-h3a-novel-strategy-for-the-design-of-anticancer-agents-for-extracranial-pediatric-solid-tumors-treatment-review/377788.png","ImageObject",300,407,{"name":92,"@type":93},"Oliver","Person",{"url":74,"name":95,"@type":96},"DocShare","Organization","application/pdf","2026-10-24","2026-09-24",true,{"@type":102,"interactionType":103,"userInteractionCount":105},"InteractionCounter",{"@type":104},"ViewAction",14,{"@type":107,"mainEntity":108},"FAQPage",[109,115,119],{"name":110,"@type":111,"acceptedAnswer":112},"What is B7-H3 and why is it considered a target for pediatric solid tumors?","Question",{"text":113,"@type":114},"B7-H3 is a checkpoint molecule that is highly expressed in extracranial pediatric solid tumors but absent or very low in normal tissues. B7-H3 is highly expressed in extracranial PSTs such as neuroblastoma, rhabdomyosarcoma, nephroblastoma, osteosarcoma, and Ewing sarcoma, whereas its expression is absent or very low in normal tissues and organs. The influence of B7-H3 on the biological behavior of malignant solid neoplasms of childhood is expressed through different molecular mechanisms, including stimulation of immune evasion and tumor invasion, and cellcycle disruption. It has been shown that B7-H3 knockdown decreased tumor cell proliferation and migration, suppressed tumor growth, and enhanced anti-tumor immune response in some pediatric solid cancers. Antibody-drug conjugates targeting B7-H3 exhibited profound anti-tumor effects against preclinical models of pediatric solid malignancies. Moreover, B7-H3-targeting chimeric antigen receptor (CAR)-T cells demonstrated significant in vivo activity against different xenograft models of neuroblastoma, Ewing sarcoma, and osteosarcoma. Finally, clinical studies demonstrated the potent anti-tumor activity of B7-H3-targeting antibody-radioimmunoconjugates in metastatic neuroblastoma. This review summarizes the established data from various PST-related st","cbCairZsdUS4hVYq","https://ap.wps.com/l/cbCairZsdUS4hVYq","pdf",387896,"English","# Abstract\n# Keywords\n# Introduction\n## Pediatric solid tumors and key examples\n## Limits of current therapies\n## Rationale for B7-H3-targeted immunotherapy","[{\"question\":\"What is B7-H3 and why is it considered a target for pediatric solid tumors?\",\"answer\":\"B7-H3 is a checkpoint molecule that is highly expressed in extracranial pediatric solid tumors but absent or very low in normal tissues.
