STUDIES ON THE EFFECT OF ADRENAL ONTHE BIOASSAY OF ANDROGEN\n\nJUN SERIZAWA \nShinagawa Plant,Sankyo Co.,Ltd.,Nishi-Shinagava,Shinagazwa-ku,Tokyo \nBioassay of androgen relies on the increase in weight of seminal vesicle and pro-state gland in a castrated rat.Eisenberg and Gordan (1950)used the levator animethod for the measurement of protein anabolic effect. These methods all utilizethe direct action of androgen on each target organ atrophied by castration. \nThe most ideal method for bioassay in general is to carry out experiments underthe most sensitive physiological condition.Adrenal excretes androgen and estrogen, togecher with corticoids which have important bearing on the metabolism.Conse-quently,it was considered that the sensitivity would be more increased by castrationand removal of the adrenal at the same time than only by castration and,if thiswere true, results somewhat different from adult rats would be obtained with im-mature rats.Reaction to androgen was therefore examined in adult and immaturerats, castrated or castrated-adrenalectomized,in order to examine the effect of adrenalon the bioassay of androgen.Further, examinations were also made on the sensitivityof various organs to androgen and some new observations were gained. \n## MATERIALS AND METHOD\n\nImmaure rats Male rats of Wistar strain,24-26 days old(body weight,35-45g)were castrated(Castrated Group). On the day following castration, the adrenals on both sides of some of thecastrated rats were removed under ether anesthesia according to the method of Grollman(1941)(Castrated-Adrenalectomized Group). \nAdult rats Male rats of Wistar strain, about 70 days old(body weight,100-130g)were castrated(Castrated Group).Fifteen days after castration, the adrenals were removed from some of thecastrated rats by the same method as immature rats(Castrated-Adrenalectomized Group).TheseCastrated and Castrated-Adrenalectomized Groups were divided into 7 groups of over 5 rats in eachgroup so as to make the average body weight of each group approximately equal. \nRats were all allowed to feed,ad libitum,on a artificial diet consisting of 60%rice flour,30%powdered skim milk,5.5%corn starch,4%casein,and 0.5%vegetable oil,and a small amount ofgreen vegetables. The Castrated-Adrenalectomized Group was given a sufficient amount of 1%NaCl solution. All the rats were kept in a thermostatic room of 24°±2℃. \nPreparation of test solution and route of administration Testosterone propionate and 19-nortestosteronephenylpropionate(Durabolin,Organon)were used as the androgen. \nWhen given in divided doses, the androgen was dissolved in peanut oil and prepared into testsolutions containing 62.5,125,250,500,1,000,or 2,000r/cc.Six doses of such solutions were \ngiven to each group as subcutaneous injection on the back in a dose of 0.1 cc per day for 7 days.A Control Group was given injection of the same peanut oil alone. \nWhen the androgen was given as a single dose, test solutions were prepared as in the case ofdivided doses to contain 625,1,250,2,500,5,000,10,000,20,000,or 40,000r/cc.These seven kindsof test solutions were given to each group in a dose of 0.2cc per rat as subcutaneous injection onthe back. \nMeasurement of organ weight In the case of divided administration, rats were sacrificed on the dayfollowing the last injection, and in the case of single administration, they were killed on the 7thor 10th day after the injection.Seminal vesicle, ventral prostate, levator ani muscles, and kidneywere removed, secretion and blood were wiped off with a filter paper, and each organ was weigheddown to 0.5mg on a torsion balance. The values were converted to the weight per 100g bodyweight. \nExperimental period The experiments were carried out during August,1958,to October,1959,andresults of three experiments are summarized. \n## RESULTS\n\n(I)Changes
