organ  \u003Cbr>constituted of several intrinsic and extrinsic muscles,  \u003Cbr>inmervated by several cranial nerves.Methods:In this study,  \u003Cbr>the nerves and their branches in the tongue were  \u003Cbr>demonstrated by Sihler's stain technique.Results:The  \u003Cbr>distributions of the hypoglossal nerve in the muscles of the  \u003Cbr>tongue were visualized.The sensitive fiber contribution of the  \u003Cbr>glossopharyngeal nerve to the posterior one-third of the  \u003Cbr>tongue was followed.In addition,the course of the lingual  \u003Cbr>nerve and its branchings in the corpus towards the tip were  \u003Cbr>demonstrated.Conclusion:We think that this overlooked  \u003Cbr>stain technique should be considered when studying the  \u003Cbr>complex innervations of structures,especially in INTRODUCTION\n\nModified Sihler's staining technique is highly  \nuseful in demonstrating neuromuscularorganization in complex structures.Using thistechnique,Liu et al.successfully studied thedistribution of intramuscular nerve branches inskeletal muscles(4).Drake et al.,on the otherhand,similarly applied Sihler's stainingtechnique to define the exact branching pattern ofnerves in the posterior cricoarytenoid muscles ofcanines,and claimed it was valuable both inunderstanding laryngeal function and in newtherapies for dysfunction(6).Wu et al.processed27 human hemi-larynges with Sihler's stainingtechnique,and suggested that the communicating  \nIt has been 100 years since Charles Sihlerfirst introduced his method for staining nervespindles and more than 50 years since otherworkers reported modifications of this techniquefor identifying innervation in the kidney,uterusand ovary in humans and in the foot and leg inanimals(1-4).  \nSihler's stain renders specimens translucentwhile counterstaining the entire nerve supply;consequently,individual peripheral nervesbecome clearly visible in fine detail and traceablein their normal three-dimensional position (5).  \nnerves supply motor innervation to thethyroarytenoid muscle as a second source,andsensory innervation to the subglottic area andcricoarytenoid joint(7).  \nThere are two other techniques for studyingintramuscular nerve supply,microdissection andserial histological sections.Microdissection isuseful only in identification of the more proximalnerve branches as they enter the substance of themuscle.Tracing nerve branches deeper into amuscle is fairly difficult and differentiating finebranches from small blood vessels andconnecting tissues could well be
