This study investigated STAT1 expression, its prognostic value, and its correlation with immune status in various types of cancer. We analyzed STAT1 expression, methylation, genomic alterations, survival, immune cell infiltration, and functional pathways using the TCGA, GTEx, GEO, TIMER, CCLE, and The Human Protein Atlas (HPA) databases. STAT1 expression was significantly higher in tumor tissues than in normal tissues and correlated with poorer overall survival. It was associated with the infiltration of B cells, CD4 + T cells, CD8 + T cells, neutrophils, macrophages, and dendritic cells. Enrichment analysis linked STAT1 to cancer-related pathways, cytokinereceptor interactions, and toll-like receptor signaling. Elevated STAT1 expression is associated with poor prognosis and immune infiltration, suggesting its potential as a biomarker and therapeutic target for cancer. \nKeywords Signal Transducer and Activator of Transcription 1 · pan-cancer analysis · Immune infiltration · posttranslational modification · enrichment analysis \nAbbreviations \nACC Adrenocortical Cancer \nBLCA Bladder Cancer \nBRCA Breast Cancer \nCESC Cervical Cancer \nCHOL Bile Duct Cancer COAD Colon Cancer READ Rectal Cancer \nDLBC Large B-cell Lymphoma \nYing Wang, Yongguang Zhang and Fengmei Wang contributed equally to this work. \n􀀍 Hu Zhao [zhaohubear@163.com](zhaohubear@163.com) \n􀀍 Qingsong Sheng [qingsongsheng@126.com](qingsongsheng@126.com) \n1 Fuzong Clinical Medical College of Fujian Medical University, 900TH Hospital of the Joint Logistics Support Force, Fuzhou, China \n2 Dongfang Hospital of Xiamen University, School of Medicine, Xiamen University, 900TH Hospital of the Joint Logistics Support Force, Fuzhou, China \nESCA \nFPPP \nGBM \nLGG \nHNSC \nKICH \nKIRC \nKIRP \nLAML \nLIHC \nLUAD \nLUSC \nMESO \nOV \nPAAD \nPCPG \nPRAD \nSARC \nSKCM \nSTAD \nTGCT \nTHCA \nTHYM \nEsophageal Cancer FFPE Pilot Phase II Glioblastoma \nlower grade glioma and glioblastoma Head and Neck Cancer \nKidney Chromophobe \nKidney Clear Cell Carcinoma \nKidney Papillary Cell Carcinoma Acute Myeloid Leukemia Liver Cancer \nLung Adenocarcinoma \nLung Squamous Cell Carcinoma Mesothelioma \nOvarian Cancer \nPancreatic Cancer Pheochromocytoma & Paraganglioma Prostate Cancer \nSarcoma \nSkin cutaneous melanoma Stomach Cancer Testicular Cancer Thyroid Cancer \nThymoma \n1 3 \nUCEC \nUVM \nTCGA \nGTEx \nGO \nKEGG \nHPA \nGEO \nCCLE \nOS \nPFI \nDFI \nDSS \nCNV \nSNV \nTMER \nSTRING \nPTM \nGDSC \nCTRP \nIC50 \nBP \nCC \nMF \nEndometrioid Cancer \nOcular melanomas The Cancer Genome Atlas the genotype-tissue expression gene ontology \nKyoto Encyclopedia of Genes and Genomes \nThe Human Protein Atlas Gene Expression Omnibus Cancer Cell Line Encyclopedia Overall Survival Progression-Free Interval Disease-Free Interval Disease Specific Survival Copy Number Variation Single-Nucleotide Variation \nTumor Immune Estimation Resource search tool for the Retrieval of Interaction Gene/Proteins \nProtein post-translational modifications Genomics of Drug Sensitivity in Cancer Clinical Trials Research Platform \nhalf maximal inhibitory concentration Biological Process \nCellular Component Molecular Function \nIntroduction \nAccording to a World Health Organization report, cancer remains the second leading cause of death globally and poses a significant threat to human health","cbCaindwiVyJQueP","https://ap.wps.com/l/cbCaindwiVyJQueP","pdf",2388007,15,"English","# Introduction\n## STAT1 and JAK/STAT signaling background\n# Materials and Methods\n## Multi-omics and database sources\n# Results\n## Expression, survival association, and immune infiltration\n## Methylation and genomic alterations\n## Enriched functional pathways\n# Discussion\n## Biomarker and therapeutic implications","[{\"question\":\"What is the main focus of this study on STAT1 in pan-cancer?\",\"answer\":\"The study examines STAT1 expression across cancer types and evaluates its prognostic value and relationship with immune status.\"},{\"question\":\"Which datasets and tools were used to analyze STAT1?\",\"answer\":\"The analysis used TCGA, GTEx, GEO, TIMER, CCLE, and The Human Protein Atlas, integrating expression, methylation, genomic alterations, survival, immune infiltration, and pathways.\"},{\"question\":\"How does STAT1 expression relate to survival and immune infiltration?\",\"answer\":\"STAT1 expression is higher in tumor tissues than in normal tissues and correlates with poorer overall survival, with associations to infiltration of multiple immune cell types.\"}]","Signal Transduction and Transcription Activator 1 in Pan-Cancer - Prognostic and Immunological Value | PDF",1790110730,38]
