Accumulating evidence links MZB1 to the regulation of intracellular Ca2⁺ signaling, integrin activation, and ER stress responses, which could influence innate-like B-cell functions and the maintenance of immune homeostasis. Aberrant expression of MZB1 has been implicated in the development of autoimmune diseases such as systemic lupus erythematosus (SLE) and rheumatoid arthritis (RA), as well as in tumor progression, including hepatocellular carcinoma and multiple myeloma. In certain cancer, MZB1 expression is associated alterationsin the tumor microenvironment that may be permissive for tumor progression. Conversely, its ability to enhance antibody responses suggests potential applications in immunotherapy. This review integrates current insights into MZB1 biology, summarizes its role in disease pathogenesis, and discusses future directions for therapeutic targeting. \nKeywords MZB1 · Immunoglobulin assembly · Immunity · Cancer-related biomarker · Therapeutic target \nAbbreviations \nMZB1 Marginal Zone B and B-1 Cell-Specific Protein \nER Endoplasmic Reticulum \nSLE Systemic Lupus Erythematosus \nRA Rheumatoid Arthritis \nIg Immunoglobulin \nNFAT Nuclear Factor of Activated T Cells \nmiRNA MicroRNA \nYue Ding, Mingyue Zhu, Ru Zhang are authors contributed equally to this work and share first authorship. \n􀀍 Fangyuan Chang [2113331472@stmail.ntu.edu.cn](2113331472@stmail.ntu.edu.cn) \nGuxin Zhou \n[13151355136@163.com](13151355136@163.com) \n1 School of Nursing and Midwifery, Jiangsu College of Nursing, Huai’an 223005, China \n2 Department of Obstetrics and Gynecology, Affiliated Dongtai Hospital of Nantong University, KANGFU West Road, Dongtai 224200, China \n3 Department of Obstetrics and Gynecology, People’s Hospital of Nanjing Medical University, Affiliated Huai’an No, Huai’an 223300, China \n3′-UTRs TCRNSCLC \nUPR \nMAT \nPBMCs \nIBD \nUC \nMS \nADA \nSSc \nPCs \nCRC \nHCC \nOC \nADCccRCCMTSCC \nTregs AMI ox-LDL CLAD HVSMCs \n3′-Untranslated Region T-Cell Receptor \nNon-Small Cell Lung Cancer Unfolded Protein Response Mesenteric Adipose Tissue Peripheral Blood Mononuclear Cells Inflammatory Bowel Disease \nUlcerative Colitis Multiple Sclerosis \nAnti-Drug Antibody Systemic Sclerosis Pathogenic Plasma Cells Colorectal Cancer Hepatocellular Carcinoma Ovarian Cancer Adenocarcinoma \nClear Cell Renal Cell Carcinoma Mucinous Tubular and Spindle Cell Carcinoma \nRegulatory T Cells \nAcute Myocardial Infarction Oxidized Low-Density Lipoprotein Chronic Lung Allograft Dysfunction Human Vascular Smooth Muscle Cells \n1 3 \nIntroduction \nMarginal zone B and B-1 cell-specific protein (MZB1), also known as pERp1 or CNPY5, is an ER-resident chaperone belonging to the CNPY family [1] . It is constitutively and abundantly expressed in innate-like B lymphocytes, particularly splenic marginal zone B cells and peritoneal B-1 cells, and is further upregulated during plasma cell differentiation [2] . In the ER, MZB1 acts as a co-chaperone by interacting with major chaperones such as BiP and GRP94 [3] . It plays an important role in the folding and assembly of immunoglobulin (Ig) subunits, thereby facilitating the secretion of polymeric IgM [4] . Recent studies also suggest that MZB1 contributes to the dimerization of J-chain-containing IgA, which is required for its transcytosis across mucosal epithelia, especially under inflammatory conditions [3, 5] . \nBeyond canonical antibody ass","cbCainJN6qtPoCb0","https://ap.wps.com/l/cbCainJN6qtPoCb0","pdf",1306905,15,"English","# Introduction\n## Core functions of MZB1 in innate-like B cells\n# Disease relevance and future directions","[{\"question\":\"What role does MZB1 play in immunoglobulin biology?\",\"answer\":\"MZB1 functions as an ER-resident chaperone that facilitates immunoglobulin subunit folding and assembly, promoting secretion of polymeric IgM and contributing to IgA dimerization for transcytosis.\"},{\"question\":\"How is MZB1 connected to immune signaling and cell function?\",\"answer\":\"MZB1 influences intracellular Ca2+ homeostasis, impacts NFAT-related downstream effectors, and promotes integrin activation through interactions such as with ERp57, supporting early immune defense.\"},{\"question\":\"Why is MZB1 considered in disease pathogenesis and therapy?\",\"answer\":\"Aberrant MZB1 expression is linked to autoimmune diseases (e.g., SLE and RA) and to tumor progression, while its ability to enhance antibody responses suggests potential immunotherapy and diagnostic/therapeutic targeting value.\"}]","Role of MZB1 in disease pathogenesis - current insights and future directions | PDF",1790693831,38]
