This study evaluates how the deubiquitinase JOSD2 influences ESCC progression by integrating bioinformatic analyses of expression, prognosis, and pathway enrichment with lentiviral manipulation of JOSD2 in KYSE30 and KYSE150 cells. Functional assays assess proliferation, colony formation, drug sensitivity, migration, invasion, and in vivo xenograft growth. Results show JOSD2 elevation correlates with poor prognosis, promotes malignant phenotypes, and increases drug resistance, partially through activation of MAPK/ERK and PI3K/AKT signaling, with interacting substrate proteins identified by mass spectrometry.",{"@graph":14,"@context":73},[15,34,56],{"@type":16,"itemListElement":17},"BreadcrumbList",[18,23,27,31],{"item":19,"name":20,"@type":21,"position":22},"https://docshare.wps.com","Home","ListItem",1,{"item":24,"name":25,"@type":21,"position":26},"https://docshare.wps.com/document/","Document",2,{"item":28,"name":29,"@type":21,"position":30},"https://docshare.wps.com/document/healthcare/","Healthcare",3,{"item":32,"name":10,"@type":21,"position":33},"https://docshare.wps.com/document/role-of-deubiquitinase-josd2-in-the-pathogenesis-of-esophageal-squamous-cell-carcinoma/337539/",4,{"url":32,"name":10,"@type":35,"image":36,"author":41,"headline":10,"publisher":44,"fileFormat":47,"inLanguage":8,"description":12,"dateModified":48,"datePublished":49,"encodingFormat":47,"isAccessibleForFree":50,"interactionStatistic":51},"DigitalDocument",{"url":37,"@type":38,"width":39,"height":40},"https://docshare.wps.com/thumbnails/role-of-deubiquitinase-josd2-in-the-pathogenesis-of-esophageal-squamous-cell-carcinoma/337539.png","ImageObject",300,407,{"name":42,"@type":43},"Finn","Person",{"url":19,"name":45,"@type":46},"DocShare","Organization","application/pdf","2026-10-28","2026-09-21",true,{"@type":52,"interactionType":53,"userInteractionCount":55},"InteractionCounter",{"@type":54},"ViewAction",9,{"@type":57,"mainEntity":58},"FAQPage",[59,65,69],{"name":60,"@type":61,"acceptedAnswer":62},"What is the main purpose of this study on JOSD2 and ESCC?","Question",{"text":63,"@type":64},"To investigate how JOSD2 affects the progression of esophageal squamous cell carcinoma (ESCC).","Answer",{"name":66,"@type":61,"acceptedAnswer":67},"How was JOSD2 manipulated and tested in ESCC cells?",{"text":68,"@type":64},"Lentiviral transduction was used to alter JOSD2 expression in ESCC cell lines KYSE30 and KYSE150, followed by assays for proliferation, colony formation, drug sensitivity, migration, and invasion.",{"name":70,"@type":61,"acceptedAnswer":71},"What mechanisms and interacting proteins are linked to JOSD2’s effects?",{"text":72,"@type":64},"Mechanistic analysis suggests JOSD2 activates the MAPK/ERK and PI3K/AKT pathways. Deubiquitinases (DUBs) have been confirmed to play a crucial role in the development of malignant tumors. JOSD2 is a DUB involved in controlling protein deubiquitination and influencing critical cellular processes in cancer. \nAIM \nTo investigate the impact of JOSD2 on the progression of ESCC. \nMETHODS \nBioinformatic analyses were employed to explore the expression, prognosis, and enriched pathways associated with JOSD2 in ESCC. Lentiviral transduction was utilized to manipulate JOSD2 expression in ESCC cell lines (KYSE30 and \n WJG [https://www.wjgnet.com](https://www.wjgnet.com) 565 February 14, 2024 Volume 30 Issue 6 \nWang WP et al. Functional assays, including cell proliferation, colony formation, drug sensitivity, migration, and invasion, were performed, revealing the impact of JOSD2 on ESCC cell lines. JOSD2's role in xenograft tumor growth and drug sensitivity in vivo was also assessed. The proteins that interacted with JOSD2 were identified using mass spectrometry. \nRESULTS \nPreliminary research indicated that JOSD2 was highly expressed in ESCC tissues, which was associated with poor prognosis. Further analysis demonstrated that JOSD2 was upregulated in ESCC cell lines compared to normal esophageal cells.
