Resveratrol (RES), a natural polyphenolic compound, has demonstrated antitumor activity in multiple cancer types; however, its underlying mechanisms in pancreatic cancer remain incompletely understood. In this study, we investigated the effects of RES on pancreatic cancer cell proliferation, migration, invasion, and epithelial–mesenchymal transition (EMT), with a particular focus on the role of inhibitor of growth family member 5 (ING5) . Pancreatic cancer cell lines PANC1 and SW1990 were treated with RES, and cell viability, clonogenic growth, migration, and invasion were assessed using CCK-8, colony formation, wound-healing, and Transwell assays, respectively. \nING5 expression and localization were evaluated by immunofluorescence, RT-qPCR, and Western blotting, and its functional significance was further examined using siRNA-mediated knockdown. \nRES treatment significantly suppressed pancreatic cancer cell proliferation, migration, invasion, and EMT in vitro, accompanied by upregulation of ING5 expression and increased E-cadherin levels with concomitant reduction of N-cadherin expression. Silencing ING5 enhanced malignant cellular behaviors and partially reversed the inhibitory effects of RES on EMT-associated phenotypes and cell growth. These findings indicate that ING5 contributes to the antitumor effects of RES and functions as an important mediator in the suppression of pancreatic cancer progression. Collectively, our results suggest that RES restrains pancreatic cancer cell aggressiveness at least in part through upregulation of ING5, highlighting the RES–ING5 axis as a potential therapeutic target for pancreatic cancer intervention. \nKeywords Resveratrol, Pancreatic cancer, ING5, Migration, Invasion, Epithelial–mesenchymal transition \nAbbreviations \nRES Resveratrol \nING5 Inhibitor Of Growth Family Member 5 PC Pancreatic cancer \nPDAC Pancreatic adenocarcinoma \nEMT Epithelial-mesenchymal transition \nSD standard deviation \nPancreatic cancer (PC), predominantly pancreatic ductal adenocarcinoma (PDAC), is the fourth leading cause of cancer-related death worldwide, owing to its increasing incidence and poor prognosis1. PDAC is a highly lethal solid malignancy, with a 5-year survival rate of approximately 10%2. Due to its anatomical location, pancreatic cancer often presents without obvious symptoms at an early stage3. Consequently, most patients are diagnosed at an advanced stage, when the disease is unresectable because of early metastasis. Cancer metastasis arises from complex interactions between intrinsic tumor cell properties and the tumor microenvironment4. However, the molecular mechanisms driving pancreatic cancer progression remain incompletely understood. Pancreatic cancer therefore represents a major global health challenge and one of the deadliest malignancies. \nTumor suppressor proteins are negative regulators of cell growth that inhibit neoplastic transformation5. The inhibitor of growth (ING) family comprises five human proteins (ING1–ING5) and three homologs in Saccharomyces cerevisiae (Yng1, Yng2, and Pho23)6. ING proteins function as essential growth regulators and \n1Department of Hepatobiliary Surgery, The First Affiliated Hospital of Xi’an Jiaotong University, 277 West Yanta Road, Xi’an 710061, China. 2Department of Hepatobiliary Surgery, Affiliated Hospital of Shaanxi University of Chinese Medicine, Xianyang 712000, China. 3Baoji Hospital of Traditional Chinese Medicine, Baoji 721000, China. \n4GuotaiWang and Yuan Yuan contributed equally to this work. 􀀍 [email: jianghe1981@163.com](email: jianghe1981@163.com) \n[www. nature.com/scientif","cbCaioYt86SNDDru","https://ap.wps.com/l/cbCaioYt86SNDDru","pdf",6075914,"English","# Background\n## Pancreatic ductal adenocarcinoma and clinical challenge\n## ING family and ING5 in cancer biology\n## Resveratrol and links to EMT\n# Study Design and Methods\n## Cell assays for proliferation, migration, and invasion\n## ING5 expression, localization, and knockdown\n# Results\n## RES suppresses proliferation, migration, invasion, and EMT\n## ING5 upregulation and EMT marker changes\n## ING5 silencing reverses RES inhibitory effects","[{\"question\":\"What effects does resveratrol have on pancreatic cancer cells in this study?\",\"answer\":\"Resveratrol significantly suppressed proliferation, migration, invasion, and EMT in pancreatic cancer cells in vitro.\"},{\"question\":\"How is ING5 involved in resveratrol’s anti-cancer effects?\",\"answer\":\"Resveratrol increased ING5 expression, and silencing ING5 enhanced malignant behaviors while partially reversing resveratrol’s inhibition of EMT and cell growth.\"},{\"question\":\"Which EMT markers changed with resveratrol treatment and ING5 manipulation?\",\"answer\":\"Resveratrol increased E-cadherin and reduced N-cadherin; ING5 knockdown counteracted these EMT-associated inhibitory phenotypes.\"}]","Resveratrol inhibits pancreatic cancer progression via the ING5 signaling pathway | PDF",1790126802,25]
