However, limited research about oncology has been conducted on Mediator complex subunit 8 (MED8) . The functional role and molecular mechanism of MED8 in bladder cancer (BLCA) require further systematic bioinformatic analysis and experimental validation. Analysis of The Cancer Genome Atlas pan-cancer cohort and multiple bladder cancer datasets revealed MED8 upregulation in various tumors. More importantly, MED8 expression was correlated with poor prognosis in bladder cancer patients. In vitro proliferation and migration assays demonstrated that MED8 knockdown suppressed bladder cancer cell proliferation and motility. Conversely, MED8 overexpression promoted cellular proliferation and migration. Meanwhile, the influence of MED8 on the growth of BLCA was validated in vivo. Integrated multi-omics sequencing (RNA-seq combined with Astral-DIA proteomics) identified Stearoyl-CoA desaturase (SCD) as a potential downstream effector of MED8 . Finally, rescue experiments confirmed that MED8 regulates bladder cancer cell proliferation and migration through SCD. In conclusion, this study is the first to reveal the oncogenic function and overexpression of MED8 in bladder cancer and its tumor-promoting effects are mediated through SCD regulation. These findings provide novel insights into the molecular mechanisms of bladder cancer progression and establish a theoretical foundation for targeting MED8 as a therapeutic strategy. \nKeywords Mediator complex subunit 8 · Stearoyl-CoA desaturase · Bladder cancer · Prognosis \nAbbreviations \nAstral-DIA proteomics BLCA \nDEG \nAstral Data-Independent Acquisition proteomics Bladder Cancer Differentially Expressed Gene \nHaiyu Zhang, Yuhan Liu, Weisheng Huang and Yanfeng Li contributed equally to this work. \n􀀍 Xinhui Liao [706751906@qq.com](706751906@qq.com) \n􀀍 Hongbing Mei [hbmei68@163.com](hbmei68@163.com) \n1 Department of Urology, The First Affiliated Hospital of Shenzhen University, Shenzhen Second People’s Hospital, Shenzhen, China \n2 Shantou University Medical College, Shantou, China \n3 Shenzhen Second People’s Hospital, Clinical Medicine College of Anhui Medical University, Shenzhen, China \nFDR False Discovery Rate \nEMBL-EBI EMBL’s European Bioinformatics Institute \nGEO Gene Expression Omnibus \nGO Gene Ontology \n4 Shenzhen Institute of Advanced Technology, Chinese Academy of Sciences, Shenzhen, China \n5 Shenzhen Institute of Synthetic Biology, Shenzhen Institutes of Advanced Technology, Chinese Academy of Sciences, Shenzhen, China \n6 Department of Urology, Shantou Central Hospital, Guangdong, China \n7 Shenzhen Clinical Research Center for Urology and Nephrology, Peking University Shenzhen Hospital, No. 1120Lianhua Road, Shenzhen, China \n1 3 \nGSEA \nHPA \nIHC \nKEGG \nMED8 \nMIBC \nNMIBC \nPol II qRT-PCR SCD \nTCGA \nTMA \nTURBT \nGene Set Enrichment Analysis Human Protein Atlas Immunohistochemical \nKyoto Encyclopedia of Genes and Genomes Mediator complex subunit 8 \nMuscle Invasive Bladder Cancer Non-Muscle-Invasive Bladder Cancer RNA Polymerase II \nQuantitative Reverse Transcription PCR \nStearoyl-CoA Desaturase The Cancer Genome Atlas Tissue Microarray \nTransurethral Resection of Bladder Tumor \nIntroduction \nBladder c","cbCait1gSJDaQ8PY","https://ap.wps.com/l/cbCait1gSJDaQ8PY","pdf",13494909,16,"English","# Abstract\n## Results overview\n## Mechanistic insight via multi-omics and rescue experiments","[{\"question\":\"What role does MED8 play in bladder cancer progression?\",\"answer\":\"MED8 is upregulated in bladder cancer and is associated with poor prognosis.
