This retrospective study evaluates whether m6A methylation regulators relate to EGFR mutations and their clinical significance. m6A-related regulatory factors are measured across EGFR-mutant and EGFR-wildtype NSCLC specimens using retrospective datasets and immunohistochemistry. Results show EGFR-mutant cases have higher METTL3 and ALKBH5 expression, with high levels independently linked to shorter progression-free survival.",{"@graph":14,"@context":73},[15,34,56],{"@type":16,"itemListElement":17},"BreadcrumbList",[18,23,27,31],{"item":19,"name":20,"@type":21,"position":22},"https://docshare.wps.com","Home","ListItem",1,{"item":24,"name":25,"@type":21,"position":26},"https://docshare.wps.com/document/","Document",2,{"item":28,"name":29,"@type":21,"position":30},"https://docshare.wps.com/document/healthcare/","Healthcare",3,{"item":32,"name":10,"@type":21,"position":33},"https://docshare.wps.com/document/m6a-methylation-regulators-mettl3-and-alkbh5-are-risk-factors-for-egfr-mutant-nsclc-article-abstract/370537/",4,{"url":32,"name":10,"@type":35,"image":36,"author":41,"headline":10,"publisher":44,"fileFormat":47,"inLanguage":8,"description":12,"dateModified":48,"datePublished":49,"encodingFormat":47,"isAccessibleForFree":50,"interactionStatistic":51},"DigitalDocument",{"url":37,"@type":38,"width":39,"height":40},"https://docshare.wps.com/thumbnails/m6a-methylation-regulators-mettl3-and-alkbh5-are-risk-factors-for-egfr-mutant-nsclc-article-abstract/370537.png","ImageObject",300,407,{"name":42,"@type":43},"Maya Linwood","Person",{"url":19,"name":45,"@type":46},"DocShare","Organization","application/pdf","2026-10-29","2026-09-23",true,{"@type":52,"interactionType":53,"userInteractionCount":55},"InteractionCounter",{"@type":54},"ViewAction",6,{"@type":57,"mainEntity":58},"FAQPage",[59,65,69],{"name":60,"@type":61,"acceptedAnswer":62},"What is the main question of the study?","Question",{"text":63,"@type":64},"The study examines whether m6A-related proteins, especially METTL3 and ALKBH5, are associated with EGFR mutations in NSCLC and what clinical significance they have.","Answer",{"name":66,"@type":61,"acceptedAnswer":67},"How were m6A-related regulators assessed?",{"text":68,"@type":64},"Associations were analyzed using datasets including The Cancer Genome Atlas, case collection and follow-up, together with immunohistochemistry and scoring.",{"name":70,"@type":61,"acceptedAnswer":71},"What did the study find about METTL3 and ALKBH5?",{"text":72,"@type":64},"EGFR-mutant NSCLC specimens showed significantly higher METTL3 and ALKBH5 expression than EGFR-wildtype specimens. Epidermal growth factor receptor tyrosine kinase inhibitor (EGFR-TKI) treatment is widely used clinically because the epidermal growth factor receptor (EGFR) is the main driver gene of lung cancer; however, the development of drug resistance is inevitable. Epigenetic abnormalities can also lead to tyrosine kinase inhibitor (TKI) resistance. However, the relationship between N6-methyladenosine (m6A)-related proteins and EGFR mutations in NSCLC and their clinical significance remain unclear. In this retrospective study, the expression of m6A-related regulatory factors in patients with EGFR mutations were analyzed to investigate their relationship with clinicopathological features and prognoses. \nMethods: The association between m6A-related regulatory factors and NSCLC was analyzed using data derived from The Cancer Genome Atlas, case collection, follow-up, immunohistochemistry, and scoring. \nResults: A total of 246 NSCLC specimens were examined in this study. Among these, 143 EGFR-mutant cases exhibited significantly higher expression of METTL3 and ALKBH5 compared to EGFR-wildtype specimens. The median progression-free survival time of patients with high METTL3 expression (SI >6) was 25.0 months, and that of patients with high ALKBH5 expression (SI >6) was 24.1 months in EGFR-mutant cases. High METTL3 and ALKBH5 expression levels are independent risk factors for progression-free survival in patients with EGFR mutations.
