This review highlights the complexity of interactions in the host’s defensive eicosanoid signaling network and its hijacking by pathogenic bacteria to their own advantage. Human lipoxygenases (ALOXs) and their mouse counterparts are integral elements of the innate immune system, mostly operating in the pro-inflammatory mode. Taking into account the indispensable role of inflammation in carcinogenesis, lipoxygenases have counteracting roles in this process. In addition to describing the structure-function of lipoxygenases in this review, we discuss their roles in such critical processes as cancer cell signaling, metastases, death of cancer and immune cells through ferroptosis, as well as the roles of ALOXs in carcinogenesis promoted by pathogenic infections. Finally, we discuss perspectives of novel oncotherapeutic approaches to harness lipoxygenase signaling in tumors. \nKeywords: ALOX; cancer; carcinogenesis; cystic fibrosis; lipoxygenase; inflammation; Pseudomonas aeruginosa \n1. Introduction \nIt is well established that many strains of Pseudomonas aeruginosa (PAE) produce the nonspecific lipoxygenase LoxA, and its homologs are present in some other opportunistic pathogens. \nGene expression studies have demonstrated elevated mRNA expression levels of LoxA during infection and biofilm formation [1]. LoxA contributes to PAE pathogenesis in cystic fibrosis patients by triggering lipid peroxidation-dependent cell death—ferroptosis, following the oxidation of host arachidonic acid (AA) containing phosphatidylethanolamines to 15-hydroperoxyAA-phosphatidylethanolamines in the bronchial epithelial cells of humans [2–4]. \nPro-and anti-tumorigenic roles of lipoxygenases, especially the immune cell-specific","cbCaivRsnUAhmgPk","https://ap.wps.com/l/cbCaivRsnUAhmgPk","pdf",26961299,37,"English","# Introduction\n## Overview of Molecular Evolution of Lipoxygenases\n## Structure-Functional Basis of PUFA Lipoxygenation","[{\"question\":\"How do opportunistic pathogens like Pseudomonas aeruginosa relate to cancer risk in this review?\",\"answer\":\"The review describes how infection can hijack the host eicosanoid signaling network, promoting lipid oxidation programs that influence carcinogenesis.\"},{\"question\":\"What role does the lipoxygenase LoxA play in cystic fibrosis patients infected with Pseudomonas aeruginosa?\",\"answer\":\"Elevated LoxA expression during infection and biofilm formation is linked to ferroptosis via lipid peroxidation after oxidation of host arachidonic-acid-containing phosphatidylethanolamines in bronchial epithelial cells.\"},{\"question\":\"Why are lipoxygenases considered both pro- and anti-tumorigenic?\",\"answer\":\"Lipoxygenases mainly operate in pro-inflammatory modes, but they also counteract aspects of carcinogenesis; the review discusses how specific ALOX isoforms can drive distinct outcomes.\"}]","Lipoxygenases at the Intersection of Infection and Carcinogenesis | PDF",1790028753,93]
