The structures of the new compounds were elucidated using HR-ESI-MS and 1D and 2D NMR data. The absolute configuration of compound 8 was assigned based on the comparison of the experimental and calculated ECD data. All isolated compounds were evaluated for their cytotoxicity against HepG2 cancer cell lines. Compounds 1 and 3–6 demonstrated significant antitumor activity compared to the positive control (cisplatin), with IC50 values ranging from 8 . 19 to 13 .97 μM. Furthermore, DARTS assay and LC-MS/ MS analysis were used to identify HSP90AA1 as the direct target of compound 5, and the interaction between compound 5 and HSP90AA1 was verified by molecular docking. \n1. Introduction \nMushrooms refer to macrofungi possessing discernible fruiting bodies that are visible to the unaided eye.1,2 Wild edible mushrooms have long been a significant component of various cultural diets such as the American, European, and Asian.3,4 Moreover, mushrooms have been utilized as fundamental components in folklore medicine and ethno-pharmacology due to their various health-promoting properties, including antioxidant, anti-cancer, anti-inflammatory, antimicrobial, anti-neurotoxic, and immunomodulatory activities.5–8 These beneficial effects can be attributed to the presence of bioactive compounds in mushrooms, such as ter \npenoids, sterols, alkaloids, polyphenols, and fatty acids.9,10 \nSuillus granulatus (L.) Roussel, known as the “weeping bolete”, is an edible and medicinal mushroom that belongs to the family Boletaceae, order Boletales, class Agaricomycetesand phylum Basidiomycota.11–15 It is widely distributed around the world and has been used as a model organism for investigating symbiotic relationships with pine trees.16–18 Nevertheless, there is a limited amount of data available regarding the chemical composition and bioactivity of this \naCollege of Life Sciences, Hebei Normal University, Shijiazhuang, Hebei 050024, \nChina. In this study, the chemical constituents of S. granulatus were characterized, and the anti-hepatocarcinoma activity and molecular targets of isolated compounds were explored. \n2.
