This emphasizes the need for targeted screening in high-risk groups, particularly those with familial predispositions and genetic mutations, such as BRCA1, BRCA2, and PALB2. This review highlights the sporadic nature of most PC cases and significant risk factors, including smoking, alcohol consumption, obesity, and diabetes. Advanced imaging techniques, such as Endoscopic Ultrasound (EUS) and Contrast-Enhanced Harmonic Imaging (CEH-EUS), have been discussed for their superior sensitivity in early detection. This review also explores the potential of novel biomarkers, including those found in body fluids, such as serum, plasma, urine, and bile, as well as the emerging role of liquid biopsy technologies in analyzing circulating tumor DNA (ctDNA), circulating tumor cells (CTCs), and exosomes. AI-driven approaches, such as those employed in Project Felix and CancerSEEK, have been highlighted for their potential to enhance early detection through deep learning and biomarker discovery. This review underscores the importance of universal genetic testing and the integration of AI with traditional diagnostic methods to improve outcomes in high-risk individuals. Additionally, this review points to future directions in PC diagnostics, including next-generation imaging, molecular biomarkers, and personalized medicine, aiming to overcome current diagnostic challengesand improve survival rates. Ultimately, the review advocates the adoption of informatics and AI-driven strategies to enhance early detection, reduce morbidity, and save lives in the fight against pancreatic cancer. | |\n\nIntroduction \nPancreatic cancer remains one of the most formidable challenges in oncology owing to its high mortality rate and poor prognosis. Despite significant advances in cancer research and treatment, the disease’s latestage diagnosis, aggressive nature, and resistance to conventional \ntherapies render it difficult to manage [1]. It is currently the seventh leading cause of cancer-related deaths worldwide, with a five-year survival rate of approximately 12%[2]. The insidious onset of pancreatic cancer, marked by nonspecific symptoms and rapid progression, underscores the immediate need for innovative strategies for early detection and risk mitigation to enhance patient outcomes and survival rates. \n* Corresponding author at: Department of Cancer Prevention, Zhejiang Cancer Hospital, Hangzhou 310022, China, Hangzhou Institute of Medicine (HIM), Chinese Academy of Sciences, Hangzhou, 310018, China. \n** Corresponding autho","cbCairjJJwrCdmK9","https://ap.wps.com/l/cbCairjJJwrCdmK9","pdf",3264214,14,"English","# Introduction\n## Screening, diagnosis, and early detection landscape\n## Risk factors and high-risk group targeting\n## Imaging approaches for early detection\n## Biomarkers and liquid biopsy technologies\n## AI-driven methods and future directions","[{\"question\":\"What patient groups should be prioritized for targeted pancreatic cancer screening?\",\"answer\":\"High-risk groups with familial predispositions and genetic mutations, such as BRCA1, BRCA2, and PALB2, are emphasized for targeted screening.\"},{\"question\":\"Which imaging techniques are highlighted for early detection?\",\"answer\":\"Advanced imaging techniques including Endoscopic Ultrasound (EUS) and Contrast-Enhanced Harmonic Imaging (CEH-EUS) are discussed as having superior sensitivity.\"},{\"question\":\"How do biomarkers and liquid biopsy contribute to early detection?\",\"answer\":\"The review describes novel biomarkers from body fluids (serum, plasma, urine, bile) and emerging liquid biopsy approaches analyzing ctDNA, circulating tumor cells (CTCs), and exosomes.\"}]","Informatics strategies for early detection and risk mitigation in pancreatic cancer patients - Review Article | PDF",1790157099,35]
