Long noncoding RNAs (lncRNAs) are potential diagnostic biomarkers in patients with EC. \nMethods: We obtained clinical information and transcriptome data for 552 patients with EC from The Cancer Genome Atlas database. Cuproptosis-associated lncRNAs were obtained through Pearson’s correlation analysis. Univariate and multivariate Cox regression analyses were applied and a signature predicting overall survival (OS) among patients with EC was constructed. We also analyzed the tumor immune microenvironment and drug sensitivity. The results were validated by quantitative real time-polymerase chain reaction, and 5-ethynyl-20-deoxyuridine and wound-healing assays. \nResults: Seven cuproptosis-associated lncRNAs related to prognosis were screened out and a signature was constructed. OS was significantly superior in the low-risk group. In addition, patients in the low-risk group had more CD8þ T cell infiltration, a stronger type II interferon response, and greater cisplatin sensitivity. Expression levels of some of the lncRNAs were significantly increased by cuproptosis. Furthermore, silencing of lncRNA AC084117.1 significantly inhibited the proliferation and migration of EC cells. \nConclusion: We constructed a seven cuproptosis-associated lncRNA signature to predict the prognosis of patients with EC with good predictive power. \nDepartment of Gynecology and Obstetrics, The First Affiliated Hospital, Zhejiang University School of Medicine, Hangzhou City, People’s Republic of China \n*These authors contributed equally to this work. \nCorresponding author: \nJianhua Qian, Zhejiang University School of Medicine First Affiliated Hospital, No. 79 Qingchun Road, Hangzhou, Zhejiang 310003, China. \nEmail: [qianjianhua@zju.edu.cn](qianjianhua@zju.edu.cn) \nCreative Commons Non Commercial CC BY-NC: This article is distributed under the terms of the Creative \nCommons Attribution-NonCommercial 4.0 License ([https://creativecommons.org/licenses/by-nc/4.0/](https://creativecommons.org/licenses/by-nc/4.0/)) which permits non-commercial use, reproduction and distribution of the work without further permission provided the original work is attributed as specified on the SAGE and Open Access pages ([https://us.sagepub.com/en-us/nam/open-access-at-sage](https://us.sagepub.com/en-us/nam/open-access-at-sage)). \nKeywords \nEndometrial cancer, cuproptosis, long non-coding RNA, prognosis prediction, immune microenvironment, survival \nDate received: 28 May 2023; accepted: 23 October 2023 \nIntroduction \nEndometrial cancer (EC) is one of the most prevalent cancers of the female reproductive system, with about 41,700 new cases worldwide in 2020, and showing an increasing trend. 1,2 The incidence rate of EC in developed countries far exceeds that of cervical cancer, making it the most common gynecological malignant tumor and the second most common female tumor.3 The main treatment is currently surgery, possibly supplemented by radiotherapy, chemotherapy, hormone therapy, or immunotherapy, according to its histology, stage and molecular classification.4,5 Notably however, there has been no significant improvement in the survival of patients with EC since the mid- 1970s.3 \nLong non-coding RNAs (lncRNAs) are functional RNA molecules with transcripts longer than 200bp, which cannot be translated into pr","cbCaieJ6GPliSOdA","https://ap.wps.com/l/cbCaieJ6GPliSOdA","pdf",1820583,16,"English","# Abstract\n## Objective\n## Methods\n## Results\n## Conclusion\n# Introduction","[{\"question\":\"What is the study’s main goal regarding endometrial cancer?\",\"answer\":\"To identify and validate a seven cuproptosis-associated lncRNA signature that predicts the prognosis of endometrial cancer patients.\"},{\"question\":\"How were cuproptosis-associated lncRNAs selected and used in the signature?\",\"answer\":\"Cuproptosis-associated lncRNAs were obtained via Pearson’s correlation analysis, then univariate and multivariate Cox regression were used to construct the overall survival (OS) signature.\"},{\"question\":\"What outcomes did the low-risk versus high-risk groups show?\",\"answer\":\"The OS was significantly better in the low-risk group, which also showed greater CD8+ T cell infiltration, stronger type II interferon response, and higher cisplatin sensitivity.\"}]","Identification and validation of a seven cuproptosis-associated lncRNA signature to predict the prognosis of endometrial cancer | PDF",1790241461]
