Methods: NSCLC and paracancer tissues from 53 cases were analyzed by RT-qPCR and Western blot; sequences/plasmids were transfected into A549 cells and malignant phenotypes were assessed with CCK-8, EdU, Annexin V-PI, and Transwell. Glycolysis was measured by glucose use, lactate production, and glycolysis-related proteins; in vivo tumorigenesis and mechanistic assays verified the hsa_circ_0075451–RBM4 interaction. RT-qPCR or Western blot was performed to detect hsa_circ_0075451 and RBM4 . Relevant sequences or plasmids were transfected into NSCLC cell line A549, and the malignant phenotype of NSCLC cells was detected by CCK-8, EdU, AnnexinV-PI double staining assay, and Transwell. Glycolysis was assessed by glucose consumption, lactic acid production, and glycolysis-related proteins. In vivo tumorigenesis assays were performed to investigate hsa_circ_0075451 functions in NSCLC. Mechanistic studies were analyzed to verify the interaction of hsa_circ_0075451 and RBM4 . \nResults Hsa_circ_0075451 levels were elevated in NSCLC, and reducing its expression diminished cell proliferation, metastasis, glycolysis, and tumor expansion. Hsa_circ_0075451 interacted with RBM4 . RBM4 down-regulation restricted NSCLC cell proliferation, metastasis, and glycolysis. RBM4 up-regulation opposed the effects of hsa_circ_0075451 knockdown. \nConclusion Hsa_circ_0075451 promotes NSCLC proliferation, metastasis and glycolysis by interacting with RBM4 . \nKeywords Non-small cell lung cancer, Hsa_circ_ 0075451, Proliferation, Metastasis, Glycolysis \n1 Introduction \nLung cancer (LC) continues to be one of the most common cancers globally, with a strong association to risk factors such as chronic smoking, environmental exposure, and genetic variations [1]. China continues to see LC as the leading cause of cancer incidence and mortality, with about 820,000 new diagnoses and 715,000 deaths from LC in 2020 [2]. NSCLC, which constitutes approximately 85% of cases [3, 4], is the most prevalent subtype. This tumor exhibits significant heterogeneity and can be classified into histological subtypes such as adenocarcinoma, squamous cell carcinoma, and large cell carcinoma [5].
