This pan-cancer study integrates cancer and normal tissue data from TCGA and GTEx to evaluate how PPM1M expression relates to survival outcomes and immune biology. Kaplan–Meier and Cox regression estimate prognostic effects, while clusterProfiler supports functional and pathway enrichment. Sanger Box, TIMER2/CIBERSORT, and TUMor-related databases assess associations with immune checkpoints, TMB, MSI, and immune-cell infiltration. Results show higher PPM1M expression in normal tissues and strong links to prognosis and immune microenvironment features, indicating potential as an immunological biomarker and therapeutic target.",{"@graph":14,"@context":73},[15,34,56],{"@type":16
BreadcrumbList",[18,23,27,31
@type":21
https://docshare.wps.com","Home","ListItem",1
@type":21
https://docshare.wps.com/document/","Document",2
@type":21
https://docshare.wps.com/document/healthcare/","Healthcare",3
@type":21
https://docshare.wps.com/document/exploring-the-immunological-role-and-prognostic-potential-of-ppm1m-in-pan-cancer/376614/",4
@type":35
"DigitalDocument
@type":38
https://docshare.wps.com/thumbnails/exploring-the-immunological-role-and-prognostic-potential-of-ppm1m-in-pan-cancer/376614.png","ImageObject",300,407
@type":43},"Patrick","Person
@type":46},"DocShare","Organization","application/pdf","2026-10-30","2026-09-24",true,{"@type":52
InteractionCounter",{"@type":54},"ViewAction",11,{"@type":57
FAQPage",[59,65,69
@type":61
What is PPM1M and why was it investigated in pan-cancer?","Question
@type":64},"PPM1M belongs to the metal-dependent protein phosphatase family. In this study, we investigated the role of PPM1M in pan-cancer.
Methods: Samples of cancer and normal tissues were obtained from the cancer genome atlas and genotype-tissue expression. Kaplan–Meier survival curves and Cox regression were used to analyze the effect of PPM1M on prognosis. Functional and pathway enrichment analyses were performed using the R package “clusterProfiler” to explore the role of PPM1M. The Sanger Box database was used to analyze the relationship between PPM1M and tumor immune checkpoint, tumor mutational burden, and microsatellite instability. The Tumor Immune Estimation Resource 2 database and CIBERSORT method were used to analyze the relationship between PPM1M and tumor-infiltrating immune cells. Finally, the cBioPortal database was used to analyze the genomic variation in PPM1M.
Results: Among the variety of tumors, the expression of PPM1M was higher in normal tissues than in cancerous tissues. The expression of PPM1M is closely associated with patient prognosis, tumor immune checkpoint, tumor mutational burden, and microsatellite instability. PPM1M is closely associated with the infiltration of immune cells into the tumor microenvironment. In addition, PPM1M is involved in the regulation of several immune-related pathways.
Conclusion: In pan-cancer, PPM1M affects patient prognosis and may be a potential immunological biomarker. Furthermore, PPM1M may be a potential therapeutic target in tumor immunology.
Abbreviations: ACC = adrenocortical carcinoma, BLCA = bladder urothelial carcinoma, BRCA = breast invasive cancer, CESC = cervical squamous cell carcinoma and endocervical adenocarcinoma, CHOL = cholangiocarcinoma, CNS = central nervous system, CNV = copy number variation, COAD = colon adenocarcinoma, DSS = disease-specific survival, ESCA = esophageal carcinoma, ESCA = esophageal cancer, GBM = glioblastoma multiforme, GSEA = gene set enrichment analysis, GTEx = genotype-tissue expression, HNSC = head and neck squamous cell cancer, HPA = human protein atlas, ICIs = immune checkpoint inhibitors, ICP = tumor immune checkpoint, IL-6 = included interleukin-6, KICH = kidney chromophobe, KIRC = kidney renal clear cell cancer, KIRP = kidney renal papillary cell cancer, LAML = acute myeloid leukemia, LGG = lower-grade glioma, LIHC = liver hepatocellular cancer, LUAD = lung adenocarcinoma, LUSC = lung squamous cell carcinoma, MESO = mesothelioma, MSI = microsatellite instability, OS = overall survival, OV = ovarian serous cystadenocarcinoma, PAAD = pancreatic adenocarcinoma, PCPG = pheochromocytoma and paraganglioma, PD-1 = programmed cell death protein 1, PFI = progression-free interval, PPM = metal-dependent protein phosphatase, PRAD = prostate adenocarcinoma, READ = rectal adenocarcinoma, SARC = sarcoma, SKCM = skin cutaneous melanoma, STAD = stomach adenocarcinoma, TCGA = the cancer genome atlas, TGCT = testicular germ cell tumors, THCA = thyroid cancer, THYM = thymoma, TIMER2 = tumor immune estimation resource 2, TMB = tumor mutational burden, TME = tumor microenvironment, UCEC = uterine corpus endometrial cancer, UCS = uterine carcinosarcoma, UVM = uveal melanoma. |
| --- |
| Keywords: human tumors, immune infiltration, immunotherapy, pan-cancer analysis, PPM1M, prognostic value |

RZ, LW and YZ contributed equally to this work.
The authors have no funding and conflicts of interest to disclose.
