The study details STRO-002 discovery and optimization, including conjugation of a cleavable 3-aminophenyl hemiasterlin linker-warhead (SC239) to an anti-FolRa antibody, yielding a homogeneous ADC with DAR 4. STRO-002 binds with high affinity, rapidly internalizes, releases the tubulin-targeting payload SC209, shows limited non-specific toxicity, demonstrates bystander killing in coculture, and remains stable in circulation with a 6.4-day mouse half-life. In xenograft and patient-derived xenograft models, a single dose strongly inhibits tumors, and combination with carboplatin or Avastin enhances efficacy. Phase I dose escalation is ongoing for ovarian and endometrial cancer patients.",{"@graph":14,"@context":73},[15,34,56],{"@type":16,"itemListElement":17},"BreadcrumbList",[18,23,27,31],{"item":19,"name":20,"@type":21,"position":22},"https://docshare.wps.com","Home","ListItem",1,{"item":24,"name":25,"@type":21,"position":26},"https://docshare.wps.com/document/","Document",2,{"item":28,"name":29,"@type":21,"position":30},"https://docshare.wps.com/document/healthcare/","Healthcare",3,{"item":32,"name":10,"@type":21,"position":33},"https://docshare.wps.com/document/discovery-of-stro-002-a-novel-homogeneous-adc-targeting-folate-receptor-alpha-for-the-treatment-of-ovarian-and-endometrial-cancers/378620/",4,{"url":32,"name":10,"@type":35,"image":36,"author":41,"headline":10,"publisher":44,"fileFormat":47,"inLanguage":8,"description":12,"dateModified":48,"datePublished":49,"encodingFormat":47,"isAccessibleForFree":50,"interactionStatistic":51},"DigitalDocument",{"url":37,"@type":38,"width":39,"height":40},"https://docshare.wps.com/thumbnails/discovery-of-stro-002-a-novel-homogeneous-adc-targeting-folate-receptor-alpha-for-the-treatment-of-ovarian-and-endometrial-cancers/378620.png","ImageObject",300,407,{"name":42,"@type":43},"hdkawk","Person",{"url":19,"name":45,"@type":46},"DocShare","Organization","application/pdf","2026-11-02","2026-09-24",true,{"@type":52,"interactionType":53,"userInteractionCount":55},"InteractionCounter",{"@type":54},"ViewAction",10,{"@type":57,"mainEntity":58},"FAQPage",[59,65,69],{"name":60,"@type":61,"acceptedAnswer":62},"What is STRO-002 and what target does it recognize?","Question",{"text":63,"@type":64},"STRO-002 is a folate receptor alpha (FolRa) targeting antibody-drug conjugate (ADC). Hallam1 \n| \u003Cbr>ABSTRACT | |\n| --- | --- |\n| STRO-002 is a novel homogeneous folate receptor alpha (FolRa) targeting antibody–drug conjugate (ADC) currently being investigatedin the clinic asa treatment for ovarian and endometrial cancers. Here, we describe the discovery, optimization, and antitumor properties of STRO-002. STRO-002 was generated by conjugation of a novel cleavable 3-aminophenyl hemiasterlin linker-warhead (SC239) to the nonnatural amino acid para-azidomethyl-L-phenylalanine incorporated at specific positions within a high affinity anti-FolRa antibody using Sutro’s XpressCFþ, which resulted ina homogeneous ADC with a drug–antibody ratio (DAR) of 4. STRO-002 binds to FolRa with high affinity, internalizes rapidly into target positive cells, and releases the tubulin-targeting cytotoxin 3-aminophenyl hemiasterlin (SC209). SC209 has reduced potential for drug efflux via P-glycoprotein 1 drug pump compared with other tubulin-targeting payloads. While STRO-002 lacks nonspecific cytotoxicity toward FolRa-negative cell lines, bystander killing of target negative cells was observed when cocultured with target positive cells. STRO-002 is stable in circulationwith no change in DAR for upto21days and has a half-life of 6.4 days in mice. A single dose of STRO-002 induced significant tumor growth inhibition in FolRa-expressing xenograft models and patient-derived xenograft models. In addition, combination treatment with carboplatin or Avastin further increased STRO-002 efficacy in xenograft models. The potent and specificpreclinical efficacy of STRO-002 supports clinical development of STRO-002 for treating patients with FolRa-expressing cancers, including ovarian, endometrial, and non–small cell lung cancer. Phase I dose escalation for STRO-002 is in progress in ovarian cancer and endometrial cancer patients(NCT03748186andNCT05200364). | |\n| 1Sutro Biopharma, South San Francisco, California. 2Seagen, South San Francisco, California. 3Genentech, South San Francisco, California. 4Gilead, Foster City, California. 5Twist Bioscience, South San Francisco, California. 6Engine Biosciences, San Carlos, California. 73T Biosciences, South San Francisco, California.\u003Cbr>Corresponding Author: Xiaofan Li, Sutro Biopharma, 111 Oyster Point Blvd, South San Francisco, CA 94080 .
