The cyclic nucleotide-degrading enzyme phosphodiesterase 10A (PDE10) hydrolyzes both cAMP and cGMP and is linked to key signaling pathways controlling proliferation, survival, and stem cell-like features. PDE10 overexpression correlates with poor relapse-free survival, and PDE10 inhibition with ADT-030 suppresses TNBC growth, improves paclitaxel efficacy, and modulates gene expression, pathways, and cellular processes. Overall, ADT-030 may work as monotherapy or with standard chemotherapy, supporting further translational research.
