This review explains how immune checkpoint blockade and CAR-T therapies work, and focuses on CD47 as a critical “don’t eat me” checkpoint that suppresses innate immune signaling. It covers CD47’s high expression in many cancers, its interaction with SIRPa to prevent phagocytosis, and evidence from antibody and small-molecule targeting. It also discusses clinical challenges including anemia due to CD47 presence on erythrocytes, proposing possible strategies.",{"@graph":14,"@context":73},[15,34,56],{"@type":16
BreadcrumbList",[18,23,27,31
@type":21
https://docshare.wps.com","Home","ListItem",1
@type":21
https://docshare.wps.com/document/","Document",2
@type":21
https://docshare.wps.com/document/healthcare/","Healthcare",3
@type":21
https://docshare.wps.com/document/deciphering-the-role-of-cd47-in-cancer-immunotherapy/406543/",4
@type":35
"DigitalDocument
@type":38
https://docshare.wps.com/thumbnails/deciphering-the-role-of-cd47-in-cancer-immunotherapy/406543.png","ImageObject",300,407
@type":43},"A glass of water","Person
@type":46},"DocShare","Organization","application/pdf","2026-10-22","2026-09-28",true,{"@type":52
InteractionCounter",{"@type":54},"ViewAction",5,{"@type":57
FAQPage",[59,65,69
@type":61
What is the role of CD47 in cancer immunotherapy?","Question
@type":64},"CD47 functions as a checkpoint that transmits a “don’t eat me” signal, suppressing innate immune signaling and helping cancer cells evade immune surveillance.","Answer
@type":61
How does CD47 prevent phagocytosis?
@type":64},"CD47 is highly expressed in many cancers and protects cancer cells from phagocytosis by binding to SIRPa on phagocytes.
@type":61
What challenges arise when targeting CD47, and how might they be addressed?
@type":64},"Targeting CD47 faces challenges such as anemia and related issues because CD47 is widely presented on erythrocytes; Immune checkpoint blockade and Chimeric antigen receptor (CAR)-T cell therapies have been successful in clinical trials. Cancer cells evade immune surveillance by hijacking inhibitory pathways via overexpression of checkpoint genes. The Cluster of Differentiation 47 (CD47) has emerged as a crucial checkpoint for cancer immunotherapy by working as a ‘‘don’t eat me” signal and suppressing innate immune signaling. Furthermore, CD47 is highly expressed in many cancer types to protect cancer cells from phagocytosis via binding to SIRPa on phagocytes. Targeting CD47 by either interrupting the CD47-SIRPa axis or combing with other therapies has been demonstrated as an encouraging therapeutic strategy in cancer immunotherapy. Antibodies and small molecules that target CD47 have been explored in pre-and clinical trials. However, formidable challenges such as the anemia and palate aggregation cannot be avoided because of the wide presentation of CD47 on erythrocytes.
Aim of view: This review summarizes the current knowledge on the regulation and function of CD47, and provides a new perspective for immunotherapy targeting CD47. It also highlights the clinical progress of targeting CD47 and discusses challenges and potential strategies.
Key scientific concepts of review: This review provides a comprehensive understanding of targeting CD47 in cancer immunotherapy, it also augments the concept of combination immunotherapy strategies by employing both innate and adaptive immune responses.
􀀁 2024 The Authors.
