This review analyzes how circadian disruption drives breast tumorigenesis through linked mechanisms spanning biological rhythms, circadian gene regulation, tumor microenvironment dynamics, and genetic polymorphisms. It synthesizes epidemiological findings on night shift work, jet lag, dietary patterns, and exercise, and highlights key circadian genes such as CLOCK, BMAL1, PER, and CRY as molecular therapeutic targets. The review further connects dysregulated circadian rhythms to epithelial-mesenchymal transition, chronic inflammation, and immunosuppression, promoting metastasis, while assessing polymorphisms for personalized risk and tailored treatment strategies. It concludes by outlining therapeutic approaches including chronotherapy, melatonin, and traditional Chinese medicine interventions, emphasizing improved outcomes through circadian-based strategies.",{"@graph":14,"@context":73},[15,34,56],{"@type":16,"itemListElement":17},"BreadcrumbList",[18,23,27,31],{"item":19,"name":20,"@type":21,"position":22},"https://docshare.wps.com","Home","ListItem",1,{"item":24,"name":25,"@type":21,"position":26},"https://docshare.wps.com/document/","Document",2,{"item":28,"name":29,"@type":21,"position":30},"https://docshare.wps.com/document/healthcare/","Healthcare",3,{"item":32,"name":10,"@type":21,"position":33},"https://docshare.wps.com/document/circadian-rhythms-and-breast-cancer-from-molecular-level-to-therapeutic-advancements/332461/",4,{"url":32,"name":10,"@type":35,"image":36,"author":41,"headline":10,"publisher":44,"fileFormat":47,"inLanguage":8,"description":12,"dateModified":48,"datePublished":49,"encodingFormat":47,"isAccessibleForFree":50,"interactionStatistic":51},"DigitalDocument",{"url":37,"@type":38,"width":39,"height":40},"https://docshare.wps.com/thumbnails/circadian-rhythms-and-breast-cancer-from-molecular-level-to-therapeutic-advancements/332461.png","ImageObject",300,407,{"name":42,"@type":43},"Levi","Person",{"url":19,"name":45,"@type":46},"DocShare","Organization","application/pdf","2026-10-29","2026-09-21",true,{"@type":52,"interactionType":53,"userInteractionCount":55},"InteractionCounter",{"@type":54},"ViewAction",8,{"@type":57,"mainEntity":58},"FAQPage",[59,65,69],{"name":60,"@type":61,"acceptedAnswer":62},"How do circadian rhythms influence breast cancer development?","Question",{"text":63,"@type":64},"Circadian rhythms regulate physiological processes through endogenous biological clocks, and their disruption contributes to breast tumorigenesis. This comprehensive review delves into the intricate interplay between circadian disruption and breast tumorigenesis from multifaceted perspectives, encompassing biological rhythms, circadian gene regulation, tumor microenvironment dynamics, and genetic polymorphisms. \nMethods and Results Epidemiological evidence underscores the profound impact of external factors, such as night shift work, jet lag, dietary patterns, and exercise routines, on breast cancer risk and progression through the perturbation of circadian homeostasis. The review elucidates the distinct roles of key circadian genes, including CLOCK, BMAL1, PER, and CRY, in breast cancer biology, highlighting their therapeutic potential as molecular targets. Additionally, it investigates how circadian rhythm dysregulation shapes the tumor microenvironment, fostering epithelial-mesenchymal transition, chronic inflammation, and immunosuppression, thereby promoting tumor progression and metastasis. Furthermore, the review sheds light on the association between circadian gene polymorphisms and breast cancer susceptibility, paving the way for personalized risk assessment and tailored treatment strategies. \nConclusions Importantly, it explores innovative therapeutic modalities that harness circadian rhythms, including chronotherapy, melatonin administration, and traditional Chinese medicine interventions. Overall, this comprehensive review emphasizes the critical role of circadian rhythms in the pathogenesis of breast cancer and highlights the promising prospects for the development of circadian rhythm-based interventions to enhance treatment efficacy and improve patient outcomes. \nKeywords Circadian rhythms · Breast cancer · Circadian gene regulation · Tumor microenvironment · Chronotherapy \nAbbreviations \nTNBC \nER \nPR \nVEGF \nVEGFR-2 \nOR \nTriple-negative breast cancer Negative estrogen receptor Progesterone receptor \nVascular endothelial growth factor \nVascular endothelial growth factor receptor-2 Odds ratio \nDou-Dou Li and Teng Zhou have contributed equally. \n* Guan-Lin Wu [wugl@sumhs.edu.cn](wugl@sumhs.edu.cn) \n* Guang-Rui Yang [yanggr@sumhs.edu.cn](yanggr@sumhs.edu.cn) \n1 School of Clinical Medicine, Shanghai University of Medicine and Health Sciences, Shanghai, China \n2 Department of Medical Oncology, Fudan University Shanghai Cancer Center, Department of Oncology, Shanghai Medical College, Fudan University, Shanghai, China \nROR \nCCGs \nBMAL1 \nPER \nCRY \nERR1 \nCSC \nCRD \nEMT \nJL \nLAN \nSNPs \nZT \nCOX \nPARP1 \nRetinoic acid-related orphan nuclear receptors Clock-controlled genes \nBrain and muscle ARNT-like protein 1 \nPeriod \nCryptochrome \nEstrogen-related receptor alpha Cancer stem cell \nCircadian rhythm disorder Epithelial-mesenchymal transition Chronic jetlag \nLight at night \nSingle nucleotide polymorphisms Zeitgebertime \nCyclooxygenase \nPoly ADP-ribose polymerase1 \nIntroduction \nBreast cancer is the most frequently diagnosed malignancy among female worldwide and is a heterogeneous multifactorial disease that attributes to complex interactions between genetic and environmental factors (Siegel et al. 2023 ; Nolan et al. 2023) .
