Canine lymphoma is the most common cancer in dogs and has a poor prognosis. We recently found that the endocytosis inhibitor dynasore suppresses the viability of human cancer cell lines, especially hematopoietic cancers, by inducing apoptosis. In the present study, we examined the anticancer effects of dynasore on five previously established canine lymphoma cell lines (CLBL- 1, Ema, Nody-1, CLC, and GL-1) . Dynasore suppressed cell viability in these canine lymphoma cell lines more effectively than in human cancer cell lines. It also induced apoptosis in CLBL-1 and Ema cells but not in peripheral blood mononuclear cells in healthy dogs or in Madin-Darby canine kidney (MDCK) cells, suggesting that the ability of dynasore to induce apoptosis is cancer-specific. Furthermore, dynasore induced a DNA damage response in CLBL-1 and Ema cells, suggesting that it acts as a genotoxic agent in canine lymphoma cell lines. These findings suggest that endocytosis inhibitors may provide a new anticancer treatment for canine lymphoma. \nKEYWORDS: dog, dynamin, endocytosis, lymphoma, mechanistic/mammalian target of rapamycin complex 1 (mTORC1) \nCancer is the leading cause of death in adult dogs [2] . Lymphoma is the most common canine cancer, accounting for 24% of all neoplastic diseases (85% of hematopoietic tumors) [9] . Although chemotherapy leads to remission and prolongs survival in most cases of canine lymphoma, the development of drug resistance significantly limits the effectiveness of chemotherapy [21], and it is, therefore, desirable to develop drugs with different mechanisms of action than those of existing anticancer drugs. \nRecently, our laboratory revealed that pharmacological inhibition of endocytosis, a cellular function for the uptake of extracellular material, leads to the activation of a DNA damage response pathway and apoptosis [14], suggesting that endocytosis inhibitors may effectively inhibit the proliferation of cancer cells. Endocytosis is a process whereby a small area of the cell membrane is invaginated into the cell and pinched off by a coil-forming protein called dynamin (dynamin 1, 2, and 3). Dynasore, a drug that inhibits dynamin GTPase activity, has been shown to suppress the proliferation and migration of human cancer cell lines in vitro [3, 7, 11, 18, 19, 22] and in vivo [3, 4] . In addition, we recently observed a tendency for dynasore to be more effective in hematopoietic cancer cell lines than in epithelial cancer cell lines [14] . \nIn this study, we examined whether dynasore affects proliferation in five previously established canine lymphoma cell lines. Our results indicate that dynasore suppressed cell viability in all five cell lines. Dynasore induced apoptosis in lymphoma cells but not in peripheral blood mononuclear cells (PBMC) from healthy dogs or Madin-Darby canine kidney (MDCK) cells, a cell line derived from a normal dog kidney. Dynasore also activates ataxia telangiectasia and Rad3-related (ATR)-Chk1 DNA damage response pathways in canine lymphoma cells. These results suggested that endocytosis inhibitors may be effective anticancer treatments for canine lymphomas. \n*Correspond","cbCais221R9sOV3n","https://ap.wps.com/l/cbCais221R9sOV3n","pdf",2128682,"English","# Abstract\n# Background\n## Cancer and treatment limitations\n## Endocytosis inhibition and dynamin\n# Results\n## Effects on canine lymphoma cell lines\n## Apoptosis selectivity and DNA damage response\n# Materials and methods\n## Cell lines","[{\"question\":\"What is dynasore and how does it relate to endocytosis in this study?\",\"answer\":\"Dynasore is a drug that inhibits dynamin GTPase activity, thereby pharmacologically blocking endocytosis.
