Bones, liver, and lungs are identified as common metastasis sites, with bone the third most frequent target and bone-metastatic disease showing poor prognosis and short median survival. Existing therapies largely inhibit progression rather than achieving cure. Emphasis is placed on understanding tumor cell interactions with the bone microenvironment, summarizing mechanisms, clinical features, and emerging treatments to improve patients’ quality of life and guide future care.",{"@graph":69,"@context":122},[70,84,105],{"@type":71,"itemListElement":72},"BreadcrumbList",[73,77,79,82],{"item":74,"name":75,"@type":76,"position":8},"https://docshare.wps.com","Home","ListItem",{"item":78,"name":9,"@type":76,"position":14},"https://docshare.wps.com/document/",{"item":80,"name":35,"@type":76,"position":81},"https://docshare.wps.com/document/healthcare/",3,{"item":83,"name":65,"@type":76,"position":19},"https://docshare.wps.com/document/beyond-boundaries-unraveling-innovative-approaches-to-combat-bone-metastatic-cancers/331795/",{"url":83,"name":65,"@type":85,"image":86,"author":91,"headline":65,"publisher":94,"fileFormat":97,"inLanguage":63,"description":67,"dateModified":98,"datePublished":99,"encodingFormat":97,"isAccessibleForFree":100,"interactionStatistic":101},"DigitalDocument",{"url":87,"@type":88,"width":89,"height":90},"https://docshare.wps.com/thumbnails/beyond-boundaries-unraveling-innovative-approaches-to-combat-bone-metastatic-cancers/331795.png","ImageObject",300,407,{"name":92,"@type":93},"Jiven","Person",{"url":74,"name":95,"@type":96},"DocShare","Organization","application/pdf","2026-10-22","2026-09-21",true,{"@type":102,"interactionType":103,"userInteractionCount":52},"InteractionCounter",{"@type":104},"ViewAction",{"@type":106,"mainEntity":107},"FAQPage",[108,114,118],{"name":109,"@type":110,"acceptedAnswer":111},"Why is bone metastasis particularly challenging in cancer?","Question",{"text":112,"@type":113},"Bone metastasis is common in cancers such as prostate and breast cancer, and it leads to severe complications. Bone is the third most frequent target for spreading tumor cells among these organs and tissues. Patients with bone-metastatic cancers face a grim prognosis characterized by short median survival time. Current treatments have proven insufficient, as they can only inhibit metastasis or tumor progression within the bone tissues rather than providing a curative solution. Gaining a more profound comprehension of the interplay between tumor cells and the bone microenvironment (BME) is of utmost importance in tackling this issue. This knowledge will pave the way for developing innovative diagnostic and therapeutic approaches. This review summarizes the mechanisms underlying bone metastasis and discusses the clinical aspects of this pathologic condition. Additionally, it highlights emerging therapeutic interventions aimed at enhancing the quality of life for patients affected by bone-metastatic cancers. By synthesizing current research, this review seeks to shed light on the complexities of bone metastasis and offer insights for future advancements in patient care. \nKEYWORDS \nbone remodeling, metastasis, treatment-related neoplasms, cancers, bone microenvironment \n1 Introduction \nMetastatic tumors, unlike primary tumors, present a significant challenge in treatment and have a high mortality rate, resulting in the death of over 90% of patients (1) . Systemic therapies, including chemotherapy, targeted therapy, and immunotherapy, have emerged as the primary strategies to combat metastatic cancers, offering improved patient survival rates and enhanced quality of life (2, 3) . \nFrontiers in Endocrinology 01 [frontiersin.org](frontiersin.org) \nMetastasis involves a complex process characterized by reduced intercellular cohesion, migration of tumor cells from the primary site through lymph nodes or the bloodstream, and establishment in distant organs while evading the immune system (4) . Following the metastasis of tumor cells to secondary locations, angiogenic processes facilitate their growth and survival by providing oxygen and nutrients (5, 6) .
