Participants included cerebral thrombosis and cerebral haemorrhage patients post-stroke and healthy controls. Serum lipids and cholesterol, triglyceride, phospholipid, binding hexoses, chromatographic fractions on sephadex G-100 and bio-gel columns were analyzed. Results show limited differences in total cholesterol and triglyceride, while ultrawatersoluble versus non-watersoluble fractions differ significantly, with distinct hexose and lipid distributions.","# A study on uitrawatersoluble lipoprotein\n\nTakemichi KANAZAWA*,Masahiro IZAWA*,Koji SHIBUTANI*,Yashuko MATSUI*,Shin IZUMIYAMA*,Hiroko MURAOKA*,Hideki MORI*,Kogo ONODERA*,Hirobumi METOKI**,Ryozo KUDo**and Yoshito SHIMANAKA** \nThere are many reports¹~5)concerning relation-ships between arteriosclerosis and lipoproteins.Serum lipoproteins⁶)are a kind of medium fordissolving of lipids as cholesterol, triglyceride andphospholipid. But authors found that some lipo-proteins in serum lipoproteins have very solublecharacter in water. Therefore to study relationshipsbetween this water solubility of lipoprotein andcerebrovascular diseases, and biochemical consti-tuents of very water solube lipoprotein, the fol-lowing experiments were performed. \n## Experimental material and method\n\n1)The subjects are 20 cerebral thrombosisesand 6 cerebral haemorrhages after 2 months fromstroke and,10 healthy persons. \n2)Types of cerebrovascular disease werediagnosed by the reference to Okinaka's7)crite-rion,and the findings of the carotid angiographyand of the CT scan. \n3)Serum was obtained from each fastingpatients early in the morning. \n4)Cholesterol,triglyceride and phospholipidwere measured by the kit using enzymatic reaction. \n5)Binding hexoses were measured by theStato's method.8). \n6)Gel column chromatographies were per-formed by using pharmacia's sephadex G-100 andbio-gel p-300 or bio-gel p-2 of Bio,Rad Labora-tories. \n7)Thin layer chromatography was performedby using Merck's kieselgel G(type 60). \n8)A definition of ultrawatersoluble lipo-protein was given to lipoprotein in supernatantafter dialysis of lipoproteins against water for \n72 hours. The supernatant was separated bycentrifugation of 3000 rpm for 30 minutes.Theprecipitate was difined as nonwatersoluble lipo-protein. \n9)Ultracentrifugation was performed by theHavel's method.9) \n## Experimental results\n\n1)As shown in Figure 1&2 there are nomarked differences in serum cholesterol and tri-glyceride between healthy persons and persons withcerebral thrombosis and cerebral haemorrhage. But significant differences are found in ultrawater-soluble lipoprotein and nonwatersoluble lipo-protein among above three groups. \nAs shown in Figure 3,serum binding hexosesare higher in strokes than those in healthy persons. While although binding hexoses in the ultra-watersoluble lipoproteins are higher as well asserum binding hexoses in stroke, those in non-watersoluble lipoproteins are lower in strokecompared with those of healthy persons. \n2)Two hundred and sixty ml sera in earlymorning to study serum lipoprotein further morewere obtained from 120 healthy persons and theultrawatersoluble lipoproteins were fractionatedfrom those sera by the same above method. \n3)As shown in Figure 4,the pattern of hexosereaction on sephadex G-100 column chromato-graphy of the serum was separated into 3 peaks ofFrI,FrII and FrIII,and molecular weight is inorder of FrI>FrII>FrIII.Cholesterol and tri-glyceride were detected in only FrI.FrIII consistsof serum glucose. \n4)The patterns by sephadex G-100 columnchromatography of ultrawatersoluble lipoproteinare shown in Figure 5.The pattern of hexosereaction was separated into two fractions, namelyFrI and FrII.FrIII of serum was not found. \nFig.1
